胃舒汤对实验性胃溃疡大鼠的治疗作用及其对NF-κB信号通路的影响

摘　　要：目的:通过观察胃舒汤对实验性胃溃疡大鼠胃组织病理及胃组织白介素-2(IL-2),核转录因子(NF-κB)p65蛋白表达水平的影响,探讨其作用机制。方法:48只清洁级Wistar大鼠成功造模后,根据完全随机数字表法随机分为正常组、模型组、奥美拉唑组、胃舒汤组。自造模后第3天,正常组和模型组给予生理盐水,奥美拉唑组、胃舒汤组分别给予奥美拉唑胶囊溶液、胃舒汤水煎液灌胃,每天1次,连续灌胃13天。观察用药过程中各组大鼠一般状态、胃大体形态学和组织病理学,比较各组的胃组织病理指数,测定并比较各组胃组织IL-2、NF-κBp65蛋白的表达水平。结果:胃舒汤组和奥美拉唑组大鼠饮食、活动量、被毛等一般状态优于模型组,且体重增长速度明显大于模型组,尤以胃舒汤组更为明显。胃舒汤组和奥美拉唑组溃疡指数明显低于模型组(P<0.01),且胃舒汤组溃疡指数明显低于奥美拉唑组(P<0.01)。模型组胃组织IL-2水平及NF-κBp65蛋白表达水平显著高于正常组(P<0.01),奥美拉唑组及胃舒汤组IL-2水平及NF-кBp65水平均显著低于模型组,且胃舒汤组的IL-2及NF-κBp65水平更低于奥美拉唑组,差异均有统计学意义(P<0.01)。结论:胃舒汤对实验性胃溃疡大鼠具有较好的治疗作用,其作用机制可能与通过抑制NF-κB的活化及表达,减少炎症因子的释放有关。Objective： To observe the curative effect of Weishu tang on stomach tissue pathology, and the expression levels of interleukin- 2（IL- 2）and Nuclear factor- κB p65（NF- κB p65） of mice with experimental gastric ulcer,and discuss its mechanism of action. Methods：After successfully molding,forty- eight clean Wistar mice were divided into normal group,model group,omeprazole group and Weishu tang group,according to the completely random number table. The third day after molding,mice in normal group and model group were given the normal saline,while mice in omeprazole group and Weishu tang group were respectively given omeprazole capsules solution and Weishu tang by gavaging one time per day continuously for 13 days. During the pharmacy process,observe the general status,the morphology and the histopathology of the mice in each group and compare the stomach tissue pathological index of each group and compare the expression level of IL- 2 and NF- κBp65 of stomach tissue. Results：The general status such as eating and drinking,activity and further of mice in Weishu tang group and omeprazole group were better than that in model group,and the speed of weight growth was obviously higher than that in model group,especially for the mice in Weishu tang group. The Ulcer index in Weishu tang group and omeprazole group was obviously less than that in model group（P〈0.01）,and the ulcer index of Weishu tang group was obviously less than that in omeprazole group（P〈0.01）.The levels of stomach tissue IL- 2 and the protein expression levels of NF- κBp65 of mice in model group were obviously higher than those in normal group（P〈0.01）. The IL- 2 levels and the protein expression levels of NF- κBp65 in omeprazole group and Weishu tang group were all obviously less than those in model group（P〈0.01）. The levels of IL- 2 and NF- κBp65 in Weishu tang group were even less than those in omeprazole group（P 0.01）. Conclusion： Weishu tang has preferable therapeutic effect on the mice with experimental
