摘  要:目的探索建立小鼠瘀热内结证模型的方法及验证当归六黄汤对本证干预的影响。方法 ICR小鼠60只,随机分为正常组、模型组、中药组(当归六黄汤组)与西药组(匹多莫德组),正常组腹腔注射生理盐水,其余3组小鼠分别予腹腔注射LPS 2mg/kg(用生理盐水配制成2mg/m L);正常组和模型组给予生理盐水灌胃,中药组给予当归六黄汤颗粒,西药组给予匹多莫德灌胃(10m L/kg·d),连续给药4周,观察小鼠体重,体温、舌象、尾部瘀斑、耳廓和爪甲颜色、汗出等表征的变化,测试耳廓、尾部微循环。结果与正常组相比,模型组小鼠表现出明显汗出、烦躁、大便干燥、舌质红紫、爪甲红紫、尾部出现明显瘀斑,其体温明显升高(P<0.05);小鼠耳廓、尾部微循环PU值及体重明显降低(P<0.05),显示了瘀热内结证的特征。与模型组比较,中药组无明显的汗出、烦躁表现,大便正常,尾部无瘀斑,体温下降(P<0.05),体重升高(P<0.05)。中药组尾部循环PU值呈现升高趋势(P<0.05)。结论腹腔注射LPS所建立的瘀热内结证小鼠模型呈现出瘀热的表征,中药当归六黄汤对其表征具有一定的干预作用。Objective To explore the establishment of heat and blood stasis in sticking models of mice,and verify the influence and intervention of Danggui Liuhuang decoction on this syndrome. Methods 60 ICR mice were randomly divided into normal group,model group,TCM( Danggui Liuhuang decoction) group and western medicine( Pidotimod) group. Normal group received intraperitoneal injection,other 3groups received intraperitoneal injection of LPS by 2mg / kg( with saline formulated into 2mg / m L) to form the models. Normal group and model group were given NS,TCM group was given Danggui Liuhuang decoction,western medicine group was given pidotimod,continuously administrating for 4 weeks,observe body weight,body temperature changes in the tongue,tail petechiae,ear and firgernail color,sweating and other characterizations,test ear and tail microcirculation. Results Compared with the normal group,model group showed significantly sweating,irritability,dry stool,purple tongue,purple finger- nail and tail significant bruising,rising body temperature( P〈0. 05),mice ear profile,tail microcirculation PU value and weight decreased significantly( P〈0. 05),showing the characteristics of heat and blood stasis in sticking. Compared with the model group,TCM group showed no significant sweating and irritability performance,normal stool,no tail bruising,decreased temperature( P〈0. 05),increased weight( P〈0. 05). TCM group showed an increasing trend in PU value of tail loop( P〈0. 05). Conclusion Danggui Liuhuang decoction has some intervention on the characterization of heat and blood stasis in sticking mice models introduced by in traperitonaeal injection of LPS.
