摘  要:试验旨在建立鸡毒支原体人工感染动物模型,为鸡毒支原体致病的分子机制研究提供基础。首先在7日龄时使用氢化可的松建立鸡的免疫低下模型,15日龄时在左右胸气囊分别注射0.5 m L浓度为1×109CCU/m L的鸡毒支原体R株菌液,观察鸡的临床症状并进行剖检,采集咽拭子、气囊和肺进行PCR检测,采用血清平板凝集试验(SPA)检测血清抗体,对气管和肺进行光镜检查。结果显示,攻毒4 d后,鸡表现出明显的呼吸道症状,解剖发现气囊浑浊、增厚并有黄色干酪样渗出物,PCR和SPA检测呈阳性,光镜下观察到气管中纤毛脱落,上皮细胞增生,黏膜下层淋巴细胞浸润,肺出血并有炎性细胞渗出。表明鸡毒支原体感染病理模型造模成功。This experiment was to develop a model for Mycoplasma gallisepticum(MG)in chicken,which could provide a basis for investigating the pathogenesis of MG. Firstly,we developed a immunocompromised model in chicken at 7 days of age with Hydrocortisone(HC),then at 15 days of age,0.5 m L MG R strain bacteria solution was injected in to the left and right air sacs with the concentration of 1×109CCU/mL. Then the clinical symptoms were observed and they were necropsied. The pharynx swab,air sac and lung were detected by PCR. The antibody in serum was tested by serum plate agglutination(SPA)test,and the pathological changes of trachea and lung tissue were analyzed by light microscope(LM).After 4 days of inoculation,the chickens showed obvious sickness in respiratory tract. The air sacs became opacificated and thicker,even yellow cheese-like exudate was found. The results of PCR detection and SPA test were positive. By LM,we could find that the cilia in tracheal was off,epithelial cells were proliferated,lymphocytes were infiltrated in the submucosa,the lung was hemorrhaged and inflammatory cells were infiltrated. The results showed that the MG infection model in chicken was successfully established.
