摘  要:目的:通过对土鳖虫不同酶解产物进行体外抗凝药效学实验,筛选强活性部位。方法:以土鳖虫不同酶解产物的得膏率、总肽含量、抗凝活力为指标,以冷冻干燥将不同酶解产物制成冻干粉,利用体外溶血块实验、血液黏度测定比较其抗凝活力。结果:仿生酶解法(胃蛋白酶+胰蛋白酶)产物的得膏率41.84%、总肽含量为26.22 mg·m L^(-1)、其抗凝活力为2.9 U·m L^(-1),其体外溶血率最高,血液黏度最低。结论:与单一酶进行酶解相比,仿生酶解法所得酶解产物具有更强的抗凝活力。Objective: To screen the active fraction by pharmacological experiment with anti- thrombogenicity on different enzymatic hydrolysates of Eupolyphaga. Methods: With the extraction rate,the total content of peptide and anticoagulant activity as indexes,different enzymatic hydrolysates were made to be lyophilized powder by freeze- drying. Comparisons of anticoagulant activities were made in vitro by hemolysis test and blood viscosity measurement. Results: The yield from bionic enzymatic method( pepsin + trypsin) was 41. 84% and the total peptide content was 26. 22 mg·m L-1and the anticoagulant activity 2. 9 U·m L-1and this results had the highest hemolysis rate in vitro and the lowest blood viscosity. Conclusion: Compared with the single enzyme digestion,the yield from bionic enzymatic method has greater anticoagulant activity.
