摘  要:目的:探讨中药复方固本防哮饮对哮喘缓解期小鼠IκB激酶(IKKβ)及核转录因子(NF-κB)表达的影响。方法:采用卵蛋白(OVA)致敏和呼吸道合胞病毒(RSV)滴鼻诱导激发BALB/c雌性小鼠,建立哮喘缓解期动物模型,随机分为正常组,模型组,固本防哮饮低、中、高剂量组,孟鲁司特钠组及地塞米松组。用Real-time PCR和Western Blot法分别检测肺组织中IKKβ、NF-κBp65 mRNA和蛋白的表达水平。结果:模型组小鼠肺组织中IKKβ、NF-κBp65 mRNA和蛋白表达均显著高于正常组(P<0.01);固本防哮饮低、中、高剂量组,孟鲁司特钠组及地塞米松组小鼠肺组织中IKKβ、NF-κBp65 mRNA和蛋白表达均低于模型组(P<0.05)。结论:哮喘缓解期气道炎症可能与IKKβ、NF-κBp65mRNA和蛋白高表达有关。固本防哮饮在一定程度上能降低IKKβ、NF-κBp65 mRNA和蛋白高表达,提示固本防哮饮能抑制IKK/NF-κB信号通路的活性,从而减轻气道炎症,预防哮喘发作。Objective: To investigate the effects of Guben Fangxiao Decoction on IκB kinase( IKKβ) and nuclear factor( NF- κB) in mice with asthma in remission. Methods: By using ovalbumin( OVA) sensitization and respiratory syncytial virus( RSV) intranasal- induced method to establish BALB/c female mice models of asthma in remission,the mice were randomly divided into normal group,model group,low,middle,high dose of Guben Fangxiao Decoction groups,montelukast group and dexamethasone group. The Real- time PCR and Western Blot were used to detect the mRNA and protein expression levels of IKKβ and NF- κBp65 in lung tissue. Results: The mRNA and protein expression levels of IKKβ and NF- κBp65 in lung tissue in model group were significantly higher than those of the normal group( P 〈 0. 01) and the mRNA and protein expression levels of IKKβand NF- κBp65 in lung tissue in low,middle,high dose of Guben Fangxiao Decoction groups,montelukast group and dexamethasone group were lower than those of the model group( P 〈 0. 05). Conclusion: Asthma in remission airway inflammation may be associated with the high mRNA and protein expressions of IKKβ and NF- κBp65. Guben Fangxiao Decoction can reduce the high mRNA and protein expressions of IKKβ and NF- κBp65 in a certain extent,suggesting that Guben Fangxiao Decoction can inhibit the activity of the IKK/NF- κB signal pathway,thereby reducing airway inflammation and preventing the asthma attacks.
