脂联素基因单核苷酸多态性与妊娠期糖尿病发病的相关性研究

摘　　要：目的:探讨脂联素基因单核苷酸多态性与妊娠期糖尿病发病的相关性。方法:选取深圳市人民医院正常孕妇静脉血标本150例,妊娠期糖尿病(GDM)孕妇静脉血共150例,采用PCR-RFLP(聚合酶链式反应-限制性片段长度多态性)技术,测定AMP1基因SPNs位点:+45T/G和+276G/T等位基因和基因型的分布频率。结果:(1)单核苷酸多态性+45T/G:GDM组与对照组比较,基因型频率分布差异具有统计学意义(P<0.05);而G等位基因频率,GDM组高于对照组,差异有统计学意义(P<0.05)。(2)单核苷酸多态性+276G/T:基因型频率分布比较,GDM组与对照组无统计学差异(P>0.05);T和G等位基因频率,GDM组高于对照组,差异无统计学意义(P>0.05)。结论:脂联素单核苷酸多态+45T/G与GDM的发生存在一定联系;单核苷酸多态性+276G/T与GDM的发生无明显关联。Objective To investigate Association of Adiponectin Gene Single Nucleotide Polymorphism with Gestational Diabetes Mellitus. Method 150 cases of normal pregnant women in the people＇s hospital of Shenzhen were selected,and 150 cases of gestational diabetes were selected. PCR- RFLP method was used to determine the distributions of allele and genotype frequencies of SNP ＋ 45 T / G polymorphism and ＋ 276 G / T. Results（1）SNP ＋ 45 T / G： genotype frequency distribution,GDM group compared with control group,the difference was statistically significant（ P〈0. 05）; G allele frequency in GDM group was higher than the control group,the difference was statistically significant（ P〈0. 05）.（2）SNP ＋ 276 G / T： genotype frequency distribution,GDM group compared with control group,there was no statistically significant difference（ P〈0. 05）. G and t allele frequency in GDM group was higher than the control group,there was no statistically significant difference（ P〈0. 05）. Conclusion There is a certain relationship between the occurrence of ＋ 45 T/G and GDM in adiponectin single nucleotide polymorphism; The occurrence of single nucleotide polymorphism ＋ 276 G/T is not significantly correlated with the occurrence of GDM.
