摘  要:目的观察丹参酮ⅡA对梗死后心力衰竭模型鼠梗死组织中ANG-TGF-PN信号通路的干预作用。方法采用通过结扎冠状动脉(左前降支)建立的梗死后心力衰竭大鼠模型,丹参酮ⅡA用药4周后,检测血清中AngⅡ的水平;检测梗死组织中转化生长因子-β1(TGF-β1)和骨膜蛋白(PN)的表达。结果 1)心衰对照组血清中AngⅡ水平的表达较正常组显著增加(P<0.05);与心衰对照组比较,3个剂量的治疗组均显著降低(P<0.05);大、中剂量组比雷米普利组显著降低(P<0.05);而小剂量组与雷米普利组下降程度接近(P>0.05)。2)大、中剂量组TGF-β1和PN的蛋白表达被有效抑制(P<0.05),其中大剂量组与雷米普利组差异无统计学意义(P>0.05),显著低于小剂量组和心衰对照组(均P<0.05),呈剂量依赖性。3)与心衰对照组比较,大剂量、中剂量组左室重量指数、梗死范围、CVF和PVCA明显降低(P<0.05),其中大剂量组与雷米普利组无明显差异(P>0.05),显著低于小剂量组(P<0.05)。结论丹参酮ⅡA可以通过AngⅡ-TGF-β1-PN信号通路,最终降低PN蛋白的表达,延缓大鼠心梗后心衰的发展,改善心室的重构。Objective: To observe the intervention effect of sodium tanshinone ⅡA sulfonate on ANG-TGF-PN signal pathways in infarction tissue of rats with heart failure. Methods: Rat models with heart failure after myocardial infarction were established byligating left anterior descending coronary artery. After treated by sodium tanshinone Ⅱ A sulfonate for 4 weeks,serum Ang Ⅱ level and the expression of TGF-β1 and PN in Infarction tissue were be detected. Results: 1)Compared with the normal group,the level of Ang Ⅱ in serum increased significantly in the control group(P〈0.05);compared with the control group,that in the three treatment groups with different doses obviously dropped(P〈0.05);the level of high doses and middle dose group decreased significantly than that of Ramipril group(P〈0.05);and the levels of small dose group was similar to that of Ramipril group(P〈0.05). 2)The expression of TGF-β1 and PN was effectively inhibited in high doses and middle dose group(P〈0.05),and there was no significant difference between high dose group and Ramipril group(P〈0.05),but was significantly lower than that in low-dose group and the control group(P〈0.05),which showed a dose dependence.3)compared with the control group,left ventricular mass index,the infarct size,collagen volume fraction and perivascular collagen area decreased obviously in high dose and middle dose group(P〈0.05),and there was no significantly difference between the high dose group and Ramipril group(P〈0.05),but was significantly lower than that in low dose group(P〈0.05). Conclusion: Sodium tanshinone ⅡA sulfonate can reduce PN protein expression by Ang Ⅱ-TGF-β1-PN signal path,delay the development of heart failure,and improve ventricular remodeling.
