摘  要:目的研究黄芪预处理对大鼠肠系膜缺血再灌注损伤后氧化应激和炎症反应的抑制作用。方法取120只实验用大鼠按随机数字表法分为假手术组、模型组、银杏叶提取物(3.15 mg/kg)预处理组和黄芪(5、10、20 g/kg)预处理组;术前7 d开始腹腔注射给药(每日1次)。通过夹闭肠系膜上动脉后去夹恢复血流再灌注的方法制备肠系膜缺血再灌注损伤大鼠模型;再灌注2 h后,测定肠组织含水量,HE染色观察肠组织病理变化并进行损伤评分(Chiu′s氏评分);检测肠组织中抗氧化酶[超氧化物歧化酶(SOD)、GSH-Px、过氧化氢酶(CAT)]活性和丙二醛(MDA)含量;测定血清中总抗氧化能力(T-AOC)水平和一氧化氮(NO)含量;酶联免疫法(ELISA)测定肠组织中炎症细胞因子(CRP、TNF-α、IL-1β、IL-6、IL-10)和内毒素水平。结果与模型组比较,黄芪(10、20 g/kg)预处理组大鼠肠组织含水量显著降低(P<0.05或P<0.01);黄芪预处理组大鼠肠系膜病变较模型组均明显改善,其中黄芪(10、20 g/kg)预处理组Chiu′s氏评分显著降低(P<0.05或P<0.01);黄芪(10、20 g/kg)预处理组肠组织中SOD、CAT活性显著升高且MDA含量显著降低(P<0.05或P<0.01),血清中NO、CRP、TNF-α、IL-6含量和内毒素水平显著降低(P<0.05或P<0.01);其中黄芪(20 g/kg)预处理组肠组织中GSH-Px活性和血清中T-AOC含量显著升高(P<0.05),血清中cGMP、IL-10含量显著升高且IL-1β含量显著降低(P<0.05或P<0.01)。结论黄芪预处理具有抑制大鼠肠系膜缺血再灌注损伤后氧化应激和炎症反应的作用,从而对肠系膜缺血再灌注损伤起到保护作用。Objective: To investigate the inhibition of Astragalus pretreatment on oxidative stress and inflammation in intestinal ischemia-reperfusion rats. Methods: 120 expremental rats were randomly devided into sham operation group,the model group,Ginkgo biloba extract(GB,3.15 kg) pretreatment group and Astragalus(5,10,20g/kg) pretreatment groups. Severn days before surgery,the drugs were given by intraperitoneal injection,once a day. The rat models were made by clipping superior mesenteric artery;two hours later,the water content was detected;the histopathological changes of intestinal tissue was observed by HE staining and the Chiu′ s score was detected;the activity of SOD,GSH-Px,CAT and the content of MDA in intestinal tissue were detected;the level of T-AOC and the content of NO,cGMP in serum were detected;the level of CRP,TNF-α,IL-1β,IL-6,IL-10 and endotoxin in intestinal tissue were detected. Results: Compared with the model group,the water content in Astragalus(10,20 g/kg) pretreatement groups were significantly decreased(P〈0.05 or P〈0.01);the histopathological changes of intestinal tissue in Astragalus pretreatement groups were significantly improved,the Chiu′ s score in Astragalus(10,20 g/kg) pretreatement groups were significantly decreased(P〈0.05 or P〈0.01),the activity of SOD,CAT in intestinal tissue were significantly increased and the content of MDA were significantly decreased(P〈0.05 or P〈0.01),the level of NO,CRP,TNF-α,IL-6 and endotoxin in intestinal tissue of Astragalus(10,20 g/kg)pretreatement groups were significantly decreased(P〈0.05 or P〈0.01);the activity of GSH-Px and the level of T-AOC in intestinal tissue were significantly increased(P〈0.05),the content of cGMP and the level of IL-10 in serum were significantly increased(P〈0.05 or P〈0.01),IL-1β was significantly decreased(P〈0.01). Conclusion: Astragalus pretreatment has inhibitive effects on oxidative stress and inflammation in intestinal ischemia-reperfusion
