摘 要:目的研究灯盏细辛对大鼠局灶性脑缺血再灌注后血管内皮生长因子(VEGF)的影响,探讨其对脑缺血再灌注损伤的保护机制。方法 SD大鼠30只,采用随机数字表法随机分为对照组、模型组及灯盏细辛组,采用线栓willis环的方法造模。治疗用灯盏细辛尾静脉给药治疗10 d,先测量治疗前及第1、5、10日4个时点大鼠脑电图病理性棘波(EEG)出现次数及局部脑血流量(rCBF),再采用酶联免疫吸附法(ELISA)检测治疗后缺血脑组织VEGF及VEGF受体Flk-1表达水平,并采用反转录聚合酶链反应(RT-PCR)检测VEGF mRNA及Flk-1 mRNA表达,TTC染色观察脑梗区细胞,比较脑梗死百分率。结果与模型组比较,灯盏细辛组大鼠VEGF、Flk-1、VEGF mRNA及Flk-1F mRNA治疗5 d后均上调,rCBF在第5、10日明显增高,EEG明显减少,脑梗死比例明显缩小(P<0.05)。结论灯盏细辛可上调大鼠局灶性脑缺血再灌注后VEGF及VEGF受体Flk-1表达,这对减轻脑缺血再灌注后血管内皮炎症反应程度、诱导缺血区血管内皮细胞增殖及血管新生、增加缺血区rCBF、缩小脑梗死区起到至关重要的作用,可明显改善脑缺血再灌注损伤造成的血管内皮功能紊乱。Objective: To study Fleabane raising VEGF after focal cerebral ischemia and reperfusion in rats,and to explore the protective mechanism against cerebral ischemia reperfusion injury. Methods: 30 SD rats were divided into the control group,the model group and fleabane group. Willis′ circle was used to make models. Tail vein injection of Fleabane was given for 10 days. EEG and r CBF was detected before treatment and on 1st,5th and 10^(th) day. ELISA was used to detect VEGF and Flk-1,RT-PCR for VEGF-mRNA and Flk-1 mRNA. TTC was dyed to observe the cells of cerebral infarction;the percentage of cerebral infarction was compared. Results:Compared with the model group,VEGF,Flk-1,VEGF-mRNA and Flk-1F mRNA in fleabane group all rose 5 days after treatment;r CBF rose obviously on 5^(th) and 10^(th) day;EEG reduced obviously,as well as Cerebral infarction ratio(P〈0.05). Conclusion: Fleabane can raise VEGF and Flk-1 after focal cerebral ischemia and reperfusion,which can reduce the extent of vascular endothelial inflammation after cerebral ischemia and reperfusion,induce ischemic vascular endothelial cell proliferation and angiogenesis,increase r CBF in the ischemic area,play a crucial role in reducing infarct area,and significantly improve endothelial dysfunction caused by cerebral ischemiareperfusion injury.
