摘 要:目的:探讨蛭丹化瘀组方对内毒素诱导肺损伤大鼠血浆降钙素基因相关肽(CGRP)、内皮素-1(ET-1)、肿瘤坏死因子(TNF-α)的影响。方法:SD大鼠84只,雌雄各半,随机分为对照组、模型组、蛭丹化瘀组方3个剂量(33、16.5、8.25 g/kg)。大鼠气管内滴注LPS复制急性肺损伤大鼠模型,称重法检测大鼠肺含水量,放免法检测各组大鼠血浆ET、CGRP、TNF-α的变化。结果:模型组大鼠肺含水量较正常组明显升高(P<0.01),蛭丹化瘀组方(33 g/kg、16.5 g/kg)(P<0.01、P<0.05)以及阳性药地塞米松(P<0.01)可降低内毒素诱导肺损伤大鼠的肺含水量;与正常组比较,模型组大鼠血浆中内皮素(ET)、降钙素基因肽(CGRP)明显增高(P<0.01);蛭丹化瘀组方(16.5 g/kg、8.25 g/kg)和阳性药地塞米松可明显降低大鼠血浆ET含量,差异较模型组明显(P<0.01);蛭丹化瘀组方33 g/kg可明显增加血浆CGRP含量;与正常组比较,模型组大鼠血浆肿瘤坏死因子(TNF-α)明显增高(P<0.01);阳性药地塞米松及蛭丹化瘀组方(33 g/kg、16.5 g/kg、8.25 g/kg)均可明显降低大鼠血浆TNF-α含量(P<0.01)。结论:蛭丹化瘀组方方通过升高内毒素诱导肺损伤大鼠CGRP的含量,降低ET-1及TNF-α含量发挥对肺损伤大鼠的保护作用。Abstract Objective:To observe the effect of Zhi Dan Hua Yu prescriptionon CGRP,ET-1 and TNF-aof lung injury rats reduced byendotoxin. Methods : A total of 84 SD rats withhalf males and half females were divided into control group, model group, Zhi Dan Hua Yu prescription group(33 g/kg, 16. 5 g/kg,8.25 g/kg)and dexamethasone group by random, detecting the changes ofwater- content inlung injury rats' lung;utilizing radioimmunoassay to detecting changes of ET and CGRP in rat plasma. Results:Compared to control group,lung water content in model group rats were significantly increased (P 〈 0. 01 ) , Zhi Dan Hua Yu prescription (33 g/kg, 16. 5 g/kg)and dexamethasone obviously reduced water content in lunginjmy rats. The endothelin (ET)and ealcitonin gene peptide(CGRP) in rat plasmaof model group were significantly increased (P 〈 0. 01 )compared to that of control group; dexametha- sone reduced ET content( P 〈 0. 01 )and Zhi Dan Hua Yu prescription( 16.5 g/kg,8.25 g/kg)reduced ET contant apparently(P 〈 0. 01 ) ; CGRP eontant in rat plasma of Zhi Dan Hua Yuprescription( 33 g/kg)were increased significantly (P 〈 0. 01 ) ; The con- tent of tumor necrosis faetor-a(TNF-ct) was increased apparently(P 〈 0. 01 ) in comparison with that of control group ; dexametha- sone and Zhi Dan Hua Yu prescription(33 g/kg, 16.5 g/kg,8.25 g/kg)reduced TNF-ain rat plasma obviously( P 〈 0. 01 ). Con- clusion:Zhi Dan Hua Yu prescriptionprotect endotoxin-induced lung injury rats by increase CGRP content and reduce ET-1 and TNF-aeontent in rat plasma.
