摘 要:通过元素分析、红外光谱、紫外光谱、差热热重分析及电化学方法,对合成的铜配合物Cu(Hq)(SSA)_2·H_2O(Hq=8-羟基喹啉,SSA=磺基水杨酸)结构、热力学性能及电化学行为做了测试,并采用紫外吸收光谱、循环伏安法和粘度测定实验,研究了该配合物与鲱鱼精DNA的键合方式。加入DNA前后配合物的紫外光谱显示,配合物的吸收峰在加入DNA后吸光强度减小,峰位发生蓝移;根据加入DNA前后配合物的循环伏安曲线,可见配合物的氧化还原峰电流减小,式量电位发生正移;粘度实验发现DNA的相对粘度随配合物的加入而不断增大。这些实验结果表明配合物与DNA通过嵌插方式发生作用,该配合物有望成为一种新型抗癌药物。A new ternary complex of Cu(Hq) (SSA)2.H2O (Hq = 8-hydroxyquinoline, SSA = 5-sulfosalicylic acid dihydrate) was synthesized and charaterized by elemental analysis, IR spectrum, UV-Vis spectrum, TG-DTA and CV methods. Herring sperm DNA as biomolecular target, and interaction with complex was studied by spectroscopy, CV and DNA viscosity titration experiment. Results showed that the UV absorption peaks of complex was blue-shifted and the intensity was weakened with the addition of DNA, also the peak current decreased significantly and potential moved positively. Viscosity titration experiment vertified that the complex increased the relative viscosity of DNA. All of the above illustrated that the complex and DNA binded by the mode of intercalation, and the complex may be used as a potential antitumor drug.
