摘  要:猪丁型冠状病毒病(PDCo VD)是新近报道的呈现世界流行性的猪肠道病毒病。为掌握我国猪丁型冠状病毒(PDCo V)的遗传变异特征,本研究对2014年11月至2015年5月之间采集的我国3个省的10个猪场的64份样品进行了检测,并对8个PDCo V毒株的S基因全长以及其中一个毒株的全基因组序列进行了测序分析,PDCo V的检测率为23.4%,比猪流行性腹泻病毒(PEDV)的检测率17.2%要高,同时PDCo V的单独感染率为17.2%,与PEDV混合感染率是6.4%,系统进化树表明本试验中的SXD1毒株与之前我国香港报道的毒株HKU15-44和HKU15-155的同源性最近,而这些毒株可能来自于中国大陆地区,且Song等进行的回溯性检测检到了PDCo V,表明PDCo V是一种重要的猪病毒性腹泻病病原,早就存在但未被检出。S蛋白的第52位氨基酸编码基因的缺失可以作为区分我国PDCo V毒株和韩国以及美国PDCo V毒株的标记。同时PDCo V毒株基因组上的ORF1a、S基因和N基因的变异提示我国应加强PDCo V的监控,尽快研制出针对PDCo VD的疫苗。Porcine deltacoronavirus disease(PDCo VD) was a newly identified porcine enteric viral disease that were spreading worldwide.To study the genetic variation characteristic of Chinese PDCo V strains,a total of 64 samples(intestine,feces) were collected from pigs with watery diarrhea of ten farms in three provinces from Nov.2014 to May 2015 for detection.In which,the S gene of eight PDCo V strains and complete genome of one PDCo V strain were sequenced and analyzed.The positive rate of PDCo V was 23.4%,higher than that of porcine epidemic diarrhea virus 17.2%.The co-infection rate of PDCo V and PEDV was 6.3%.The phylogenetic tree analysis revealed that the strain SXD1 might be originated from the previously reported Chinese strains(HKU15-44,and HKU15-155),which could be collected from Chinese mainland pig farms.The PDCo V can be detected in Song's retrospective study.All the above showed that PDCo V was an important undiagnosed porcine enteropathogen in Chinese mainland farm.One amino acid(aa) deletion in the 52 aa of S protein can be a genetic marker to discriminate Chinese field strains with epidemic strains in USA and South Korea.The highly variable regions in ORF1 a,S gene and N gene remained us to strengthen the monitoring of PDCo V and develop a vaccine against PDCo VD as soon as possible.
