摘  要:目的探讨苯并芘[Benzo(a)pyrene,BaP]损伤小鼠胎盘绒毛组织细胞的作用机制及山奈酚(kaempferol,KA)的保护作用。方法选取性成熟ICR小鼠150只(100只雌鼠,50只雄鼠),随机分为5组,每组30只(20只雌鼠,10只雄鼠),雌雄分开饲养。A组予BaP 2 mg/(kg·d);B、C、D组予BaP 2 mg/(kg·d),同时分别予KA 100、200、400 mg/(kg·d);灌胃E组予相同体积的0.9%氯化钠注射液灌胃。15 d后雌雄鼠同笼,第2天发现精液栓者为受孕,将受孕鼠继续灌胃7 d后处死,取胎盘组织测定Bax与Bcl-2基因mRNA的表达水平及细胞色素P450(CYP450)、谷胱甘肽过氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)的活性。结果 B、C、D组Bax基因mRNA表达水平及CYP450酶活性均高于E组(P<0.05),低于A组(P<0.05),且随着KA剂量的增加表达量及活性均逐渐降低(P<0.05);B、C、D组Bcl-2基因mRNA表达水平及GSH-PX、SOD活性均低于E组(P<0.05),高于A组(P<0.05),且随着KA剂量的增加表达量及活性均逐渐升高(P<0.05)。结论 BaP通过升高Bax基因表达量与CYP450活性、降低Bcl-2基因表达量与GSH-PX、SOD活性使胎盘绒毛组织细胞凋亡,而KA通过阻止这一途径抑制其凋亡。Objective To study the mechanisms of action of injury that BaP[Benzo( a) pyrene]acts on placental villus and the effect of Kaempferol opposing BaP. Methods 150 viripotent ICR mice( 100 male,50 female mice) were randomly divided into five groups,( n = 30 every group; 20 male mice,10 female mice). Female mice and male mice were raised seperately. The mice were lavaged with BaP 2 mg /( kg·d) in group A,the mice were lavaged with BaP 2 mg /( kg·d) and KA 100,200,400 mg /( kg·d) in group B,C and D and group E was lavaged with normal saline( 0. 9%),After 15 days,male mice mated with female mice,and on the next day,gestational age was determined based on the presence of a vaginal plug,and lavaging was continued for 7 days. The mice were euthanized and the gene of Bax and Bcl-2 in the level of mRNA and the activity of cytochrome 450,GSH-PX and SOD in placental villus were detected. Results Group B,C,and D expressed more Bax than group E in the level of mRNA( P〈0. 05),Group B,C,and D expressed less Bax than group A in the level of mRNA( P〈0. 05),with the increase of KA dose,the expression of Bax depressed in the level of mRNA( P〈0. 05); Group B,C and D expressed less Bcl-2 than group E in the level of mRNA( P〈0. 05),Group B,C and D expressed more Bcl-2 than group A in the level of mRNA( P〈0. 05),with the increase of KA dose,the expression of Bcl-2 increased in the level of mRNA( P〈0. 05);the activity of cytochrome P450 in Group B,C and D were better that of Group E,lower than that of group A( P〈0. 05),with the increase of KA dose,the activity of cytochrome P450 decreased( P〈0. 05); the activity of GSH-PX,SOD in Group B,C,and D were lower than that of Group E and higher than that of group A( P〈0. 05),with the increase of KA dose,the activity of GSH-PX,SOD increased( P〈0. 05).
