摘  要:目的:研究红景天苷对缺氧条件下H9C2心肌细胞凋亡及Caspase-3表达的影响。方法:建立心肌细胞缺氧模型,本实验将细胞分为正常对照组、低氧组、低氧加红景天苷组(低剂量10mg/L、中剂量50mg/L、高剂量100mg/L),按实验设计因素处理后,用MTT检测取对数生长期细胞。TUNEL染色观察细胞凋亡,Westernblot测定Caspase-3表达。结果:与低氧组比较,红景天苷组各组均可明显提高缺氧损伤的H9C2心肌细胞存活率(P<0.01),降低细胞凋亡率(P<0.01),可有效减少缺氧H9C2心肌细胞内Caspase-3的表达(P<0.01)。结论:红景天苷可通过提高低氧条件H9C2心肌细胞存活率,降低细胞凋亡率及下调Caspase-3,从而保护心肌细胞。Objective:To study the effects of Salidrosides on the apoptosis of H9C2 cardiomyocyte and the ex- pression of Caspase - 3 under the hypoxic condition . Methods : Hypoxic culture model in vitro was established to test the apoptosis in H9C2 cells and the expression of Caspase -3. In this study, cells were divided into normal con- trol group, hypoxic group, hypoxia plus Salidrosides group. The doses of Salidrosides adding to the culture were three kinds of concentration: lowl 0mg/L; medium 50mg/L; and high 100mg/L. The cells viability was measured with MTT, using TUNEL to detect the rate of myocardial cell apoptosis;H9C2 cell apoptosis gene expression of Caspase -3 detected by Western blot after treating with experimental factors designed. Result:Compared with the hypoxic group, pretreatment with 10,50 and 100 mg/L of Salidroside could significantly improve the hypoxic injury of H9C2 cell survival rate(P 〈0.01 )and reduce the ceils apoptosis rate(P 〈0.01 ). Furthermore, Salidroside groups could effectively reduce the hypoxic injury of H9C2 Caspase -3 expression in the cells(P 〈 0.01 ). Conclusion:Salidro- side can protect the H9C2 cells from hypoxie injury and improve the H9C2 cell survival rate. At the same time, it can also reduce the the cell apoptosis rate and enhanced Caspase -3 expression.
