摘 要:目的:观察逍遥散对慢性束缚应激肝郁脾虚证焦虑模型大鼠血清炎性因子白介素(interleukin,IL-1β)、IL-6、肿瘤坏死因子-α(tumor necrosis factor,TNF-α)的影响,以此研究逍遥散的干预作用。方法:28只雄性SD大鼠随机分为正常组、模型组、逍遥散组、氟西汀组;正常组不做任何处理,模型组、逍遥散组和氟西汀组通过连续14天慢性束缚应激建立肝郁脾虚证焦虑模型,逍遥散组和氟西汀组于每天上午束缚前分别灌服逍遥散混悬液(0.006 1 g·g-1)以及氟西汀(0.002mg·g-1),正常组和模型组灌服去离子水。第15天,观察各组大鼠的宏观表征、体质量变化、新环境进食抑制实验,高架十字迷宫实验。第16天,用ELISA方法测定大鼠血清的IL-1β、IL-6、TNF-α的变化。结果:实验第15天模型组的行为学均明显变化。体质量:正常组、模型组、逍遥散组和氟西汀组分别为(353.14±5.80)g、(334.17±5.18)g、(353.95±8.00)g、(355.65±4.92)g,模型组明显低于其余3组(P<0.05);新环境进食抑制实验:正常组、模型组、逍遥散组和氟西汀组分别为(1.151±0.50)min、(3.48±0.53)min、(1.29±0.25)min、(1.85±0.25)min,模型组明显高于其余3组(P<0.05,P<0.01);高架十字迷宫实验:开放臂进入次数的正常组、模型组、逍遥散组和氟西汀组分别为(3.86±0.67)次、(1.28±0.68)次、(4.28±0.94)次、(3.86±0.85)次,开放臂停留时间的正常组、模型组、逍遥散组和氟西汀组分别为(15.46±6.72)s、(0.17±0.11)s、(12.38±4.27)s、(11.33±2.01)s,与正常组比较,开放臂进入次数与开放臂停留时间的逍遥散组及氟西汀组大鼠无统计学差异(P>0.05)。与模型组相比,除氟西汀组开放臂停留时间以外,其余3组大鼠的开放臂进入次数与开放臂停留时间均高于模型组(P<0.05);正常组IL-1β、IL-6、TNF-α分别为(28.20±7.53)ng·L-1、(26.70±8.86)ng·L-1、(67.65±6.73)ng·L-1,模型组IL-1β、IL-6、TNF-α分别为(91.50±28.70)ng·L-1Objective: This experiment was designed to measure the affect of Xiao Yao San on treating liver depression and spleen deficiency syndrome anxiety by recording the changes of inflammatory factor IL^-1β,IL-6,TNF-α levels in the blood serum in chronic immobilization stress( CIS) model rats. Methods: 28 male SD rats were randomly divided into 4 groups: normal group,model group,Xiao Yao San group and fluoxetine group. We didn't do any processing in normal group,but model,Xiao Yao San and fluoxetine groups by continuous 14 days immobilization stress induced liver depression and spleen deficiency syndrome anxiety,before every morning bondage administered Xiao Yao San( 0. 006 1 g·g- 1) to the Xiao Yao San group,fluoxetine( 0. 002 mg·g- 1) to the fluoxetine group and administered a deionized water to the normal and model groups. After 15 days,we observed macro characterization,weight change,elevated plus maze and novelty suppressed feeding tests. We used ELISA to measure IL^-1β,IL-6,TNF-αlevels in blood serum of rats after death on day 16 Results: The model group of 15 days of behavioral changes significantly. Weight:normal,model,Xiao Yao San and fluoxetine groups respectively( 353. 14 ± 5. 80) g,( 334. 17 ± 5. 18) g,( 353. 95 ± 8. 00),( 355. 65 ± 4. 92),the model group was significantly lower than the other three groups( P〈0. 05); The novelty suppressed feeding tests: normal,model,Xiao Yao San and fluoxetine groups respectively( 1. 151 ± 0. 50) min,( 3. 48 ± 0. 53) min,( 1. 29 ± 0. 25)min,( 1. 85 ± 0. 25) min,the model group was significantly higher than the other three groups( P〈0. 05,P〈0. 01); Elevated plus maze test: Open arm entry number of normal,model,Xiao Yao San and fluoxetine groups respectively( 3. 86 ± 0. 67) n,( 1. 28 ± 0. 68) n,( 4. 28 ± 0. 94) n,( 3. 86 ± 0. 85) n,open arm residence time of normal,model,Xiao Yao San and fluoxetine groups respectively( 15. 46 ± 6. 72) s,( 0. 17 ± 0. 11) s,(
