摘 要:目的:通过检测钩藤治疗小鼠急性脑缺血后组织中金属蛋白酶(metalloproteinase,MMP),并分析其对脑缺血的抑制作用和机理。方法:建立急性脑缺血小鼠模型,分为钩藤组、西药组和对照组,分别在其断头时给予钩藤、常规西药和生理盐水,记录其呼吸次数和时间。结果:钩藤对MMP-16活性抑制作用明显,质量浓度为52 mg·L-1左右;钩藤组、西药组、对照组小鼠呼吸次数分别为(18.37±3.56)次、(16.84±2.94)次、(14.62±3.18)次;钩藤组、西药组、对照组小鼠呼吸时间分别为(28.64±4.35)min、(25.91±5.49)min、(23.24±4.16)min;钩藤组、西药组、对照组小鼠在密闭瓶子中存活时间分别为(27.69±2.15)min、(26.94±3.48)min、(24.16±4.23)min;钩藤组小鼠未出现中毒和器官异常现象,且其呼吸次数、时间以及耐压缺氧能力均高于其他两组(P<0.05)。结论:钩藤对脑血管的治疗作用机制为抑制MMP活性,为临床治疗提供了重要的理论依据。Objective: Through the detection of Uncaria treatment of mice with acute cerebral ischemia tissue metalloproteinase and analyzed its on cerebral ischemia inhibition effect and mechanism. Methods: The establishment of acute cerebral ischemia mice model,divided into the Uncaria group,western medicine group and control group,given its ends respectively in uncaria,routine western medicine and saline,recorded the breathing frequency and time. Results: MMP-16 significantly inhibited the activity by Uncaria,in 52 mg · L^-1; the western medicine group,Uncaria group,control group of mice breathing respectively( 18. 37 ±3. 56),( 16. 84 ± 2. 94),( 14. 62 ± 3. 18); the RTA group the western medicine group,control group,mice breathing time respectively( 28. 64 ± 4. 35) min,( 25. 91 ± 5. 49) min,( 23. 24 ± 4. 16) min;( 27. 69 ± 2. 15) min,( 26. 94 ± 3. 48) min,( 24. 16 ±4. 23) min; Uncaria group,western medicine group,the control group of mice in the closed the bottle survival time( 27. 69 ±2. 15) min,( 26. 94 ± 3. 48) min,( 24. 16 ± 4. 23) min; Uncaria treatment group mice did not appear abnormal phenomenon of poisoning and organ and the respiratory frequency,time and pressure resisting hypoxia were higher than in the other two groups( P〈0. 05). Conclusion: The mechanism treatment of Uncaria on cerebrovascular is inhibition of MMP activity,for the clinical treatment providing an important theoretical basis.
