摘 要:目的对大鼠进行股动脉血流介导的血管舒张功能(FMD)检测,建立标准化检测方法。方法 21只同周龄Sprague-Dawley(SD)大鼠,随机分为4组:高血流速度(HFV,取血流速度≥100mm/s的血管段测量)组(n=5)、低血流速度(LFV,取血流速度<100 mm/s的血管段测量)组(n=6)、一氧化氮合酶抑制剂左旋硝基精氨酸甲酯(l-NAME)组(取血流速度≥100mm/s的血管段测量,同时注射l-NAME 8mg/kg,n=5),溶剂对照组(取血流速度≥100mm/s的血管段测量,同时注射生理盐水0.1mL/100g体质量,n=5),使用动物超声系统在血管封堵器压迫股动脉造成缺血5min后的0、60、120、180、240、300s分别测定不同时间点的FMD及血流速度,同时观察l-NAME注射对FMD的影响。结果缺血再灌注后5min内,HFV组与LFV组大鼠股动脉FMD在120s时达到峰值,高于l-NAME组FMD峰值[(16.49±2.23)%、(12.05±0.97)%比(6.30±1.04)%,均P<0.05],各组血流速度及管壁切应力(WSS)随时间增加逐渐降低(P_(趋势)<0.05)。与HFV组相比,l-NAME组WSS升高(P<0.05)。而溶剂对照组与HFV组在各项指标差异均无统计学意义。结论测定大鼠股动脉HFV段血管FMD是无创、高效而且可连续监测的一种内皮功能检测方法。Objective To establish a standard detection method of flow-mediated dilation(FMD)of femoral artery in rats. Methods A total of 21 Sprague-Dawley(SD)rats were randomly assigned into 4 groups,including high flow velocity(HFV)group(n=5,choosing segmental vessels with flow velocity above 100 mm/s),low flow velocity(LFV)group(n=6,choosing segmental vessels with flow velocity below 100 mm/s)and nitric oxide synthetase inhibitor NG-nitro-l-arginine methyl ester(l-NAME)group(n=5,choosing segmental vessels with flow velocity above 100 mm/s and injecting 8 mg/kg l-NAME),solvent control group(n=5,choosing segmental vessels with flow velocity above 100 mm/s and injecting 0.1 mL/100 g normal saline). Vascular closure device was used to compress the femoral artery of SD rats. FMD and blood flow velocity were measured by ultrasound system at 0,60,120,180,240 and 300 safter 5 minutes of the compression. Meanwhile,the influence of l-NAME on FMD was also observed. Results The FMD of femoral artery in the HFV and LFV group reached the peak at 120 safter 5 minutes of ischemia and reperfusion,and the FMDs in these two groups were larger than that in the l-NAME group[(16.49±2.23)%,(12.05±0.97)% vs(6.30±1.04)%,respectively,both P0.05]. Blood flow velocity and wall shear stress(WSS)were gradually decreased over time in all groups(P_(trend)0.05). Compared with the HFV group,the WSS in the l-NAME group was increased(P0.05).But there were no significant differences in all indicators between the solvent control group and HFV group. Conclusion The detection of FMD of femoral artery in rats was an noninvasive and effective detection method of endothelial function,and which can be continuously monitored.
