正肝方药物血清对甲胎蛋白刺激的AFPsiRNA HepG2细胞系凋亡的影响

摘　　要：目的:观察正肝方药物血清对甲胎蛋白(AFP)刺激的AFPsiRNA HepG2细胞凋亡的影响。方法:正肝方浸膏粉经灌胃给药制备正肝方大鼠血清。体外培养AFPsiRNA HepG2细胞,将细胞分为5%正常大鼠血清组、5%正肝方大鼠血清组、10%正常大鼠血清组、10%正肝方大鼠血清组、5%正常大鼠血清+AFP组、5%正肝方血清+AFP组、10%正常大鼠血清+AFP组、10%正肝方大鼠血清+AFP组共8组,流式细胞术检测细胞凋亡。结果:无AFP刺激时,正肝方大鼠血清与正常大鼠血清对HepG2细胞凋亡相比较,差异无显著性意义(P>0.05);当有AFP刺激时,正肝方血清与正常大鼠血清对细胞凋亡相比较,差异有显著性意义(P<0.01)。结论:正肝方大鼠血清能显著诱导AFPsiRNA HepG2细胞凋亡。其机制是通过影响AFP介导的肝癌细胞凋亡通路来实现的。Objective： To observe the effect of ZhengGan Recipe on cell apoptosis of AFPsiRNA-HepG2 cells stimulated by alpha fetoprotein. Methods：The drug sera was collected from rats, AFPsiRNA-HepG2 cells were cultured with drug sera and stimulated by AFP and without AFP. The cell apoptosis was detected by flow cytometry（FCM）. Results： Without AFP, there were no distinction between normal groups and drug sera groups （ P 〉 0. 05） ; but with AFP, the drug sera markedly induced the apoptosis of cells compared with normal groups （ P 〈 0. 01 ）. ZhengGan Recipe had significant effect on the apoptosis of AFPsiRNA-HepG2 cells. Conclusion： ZhengGan Recipe induced the apoptosis of hepatocarcinoma ceils via influencing the ceils apoptosis by AFP.
