甲乙煎对肝纤维化大鼠肝细胞转化生长因子-β1蛋白水平的影响

摘　　要：目的:观察中药甲乙煎对肝纤维化大鼠肝细胞TGF(转化生长因子)-β1蛋白表达水平、肝组织炎症活动度和肝纤维化程度的影响,以探讨中药甲乙煎抗肝纤维化的机理。方法:60只大鼠除正常对照组10只外其余50只依据文献进行造模,并依据要求给予正常对照组大鼠等容量的0.9%氯化钠注射液灌胃。模型组30只随机分为3组(模型对照组及甲乙煎高、低剂量组各10只),甲乙煎高、低剂量组大鼠分别给予22g·kg^(-1)·d^(-1)、5.5 g·kg^(-1)·d^(-1)甲乙煎灌胃治疗,模型组大鼠灌胃生理盐水,共4周。结果:①中药甲乙煎高、低剂量治疗后,大鼠肝组织炎症活动度和肝纤维化程度评分均明显降低,尤其是甲乙煎高剂量组大鼠肝组织炎症活动度和肝纤维化程度评分降低最明显,与甲乙煎低剂量组、模型组比较,差异有显著性意义(P<0.05);②高剂量甲乙煎能下调大鼠肝组织TGF-β1的表达,与甲乙煎低剂量组、模型组比较,差异有显著性意义(P<0.05)。结论:中药甲乙煎能通过下调TGF-β1的表达而明显控制实验性肝纤维大鼠的肝组织炎症和抑制其纤维化程度,从而起到改善肝肝纤维化的作用。Objective ：To observe the effects of Jiayijian on the expression level of TGF-β1 in the hepatocytes of rats with hepatic fibrosis, as well as its impact on the grade of inflammation and the degree of hepatic fibrosis to explore the possible mechanism of Jiayijian＇s anti-hepatic fibrosis. Methods：After successfully establishing the hepatic fibrosis rats model based on some latest research reports, the rats were divided into normal control group（10） and model group（30）. And then the 30 rats of model group were randomly divided into another 3 groups （ model control group, the low and high dosage of Jiayijian group）. The rats of the high and low dose group were given Jiayijian at dosage of 22g/kg/d and 5.5g/kg/d by gavage for 4 weeks. The normal control and model control group were given, according to requirements, the same volume of 0.9% sodium chloride injection by gavage once a day for 4 weeks as well. Results： For the low and high dosage of Jiayijian groups, the degree of rat liver tissue inflammation,fibrosis grade scores and TGFβ1 were significantly reduced after treatment. In particular, compared with the hepatic fibrosis model control group and the low dose group, the difference was significant in the high dose group （ P 〈 0.05 ）. Conclusion： Chinese medicine Jiayijian may down-regulate the expression of TGF-131, relieve the inflammation and inhibit fibrosis in hepatic tissue of experimental rats with hepatic fibrosis, and thereby play an important role in improving the degree of hepatic fibrosis.
