摘  要:[目的]利用生物技术手段提高盖杨抗寒性并扩大其栽培区域。[方法]以盖杨叶片为外植体,研究不同激素组合对盖杨叶片分化及生根的影响,建立盖杨叶片高频植株再生体系。[结果]盖杨不定芽分化最适培养基配方为MS+6-BA 0.5 mg/L+NAA 0.20 mg/L+蔗糖25 g/L+琼脂6.0 g/L,p H 6.5,不定芽再生频率为96.67%,平均分化芽数20.3个;最适生根培养基配方为1/2MS+IBA0.3 mg/L+蔗糖20 g/L+琼脂6.0 g/L,p H 6.0,生根率可达100%,平均生根数15.8条。[结论]该研究建立了盖杨高效组织培养再生系统,为该杨树品种的快速无性繁殖和基因的遗传转化提供了依据。[Objective] The aim was to improve the cold resistance and expand its cultivation area of Populus gaixianensis by means of biotechnology.[Method] Leaves of P.gaixianensis were used as explants to study the effect of different hormone combinations on leaf differentiation and rooting,and establish a high efficient plant regeneration system.[Result] The results showed that the optimum medium for adventitious bud regeneration was MS +6-BA 0.5 mg/L + NAA 0.20 mg/L + sucrose 25 g/L + agar 6.0 g/L,pH 6.5,with the frequency of adventitious bud regeneration of 96.67% and the number of shoots of 20.3;the optimum medium for rooting was 1/2MS + IBA 0.3 mg/L + sucrose 20 g/L + agar 6.0 g/L,pH 6.0,the rooting rate could reach 100%,and the average number of rooting was 15.8.[Conclusion] The study establishes high efficient tissue culture regeneration system for P.gaixianensis,lays a foundation for rapid vegetative propagation and genetic transformation of genes.
