摘　　要：目的观察CKLF1-C19多肽对小鼠特应性皮炎(AD)样皮损的影响。方法将2,4-二硝基氟苯外搽在8周龄BALB/c小鼠剃毛后的背部皮肤上,以建立实验性特应性皮炎样模型并进行药物早期干预。将小鼠分为7组:模型组(不治疗)、灭菌水组(腹腔注射灭菌水)、治疗组(C19多肽1μg,10μg,100μg腹腔注射)、氢化可的松组(腹腔注射氢化可的松)和空白对照组。建模第2天开始给药,连续5周,观察小鼠背部皮损并评分,测量皮褶厚度,并在皮损处取材,进行HE染色及甲苯胺蓝染色以观察小鼠皮损组织病理变化并计数肥大细胞。结果 C19多肽早期干预可缓解皮肤红斑、鳞屑、水肿及炎细胞浸润,在一定程度上减轻皮肤炎症反应。结论 C19多肽能缓解小鼠AD模型的炎症反应,可能与抑制炎症细胞趋化反应有关。Objective To observe the effects of CKLF1-C19 polypeptide on the skin lesions of atopic dermatitis-like murine model. Methods A model of atopic dermatitis was established in 8-week-old BALB/c mice by repeatedly applying 2% 2, 4-dinitrofluorobenzene （DNFB） to shaved dorsal skin, and an early pharmacological intervention was also needed. The target mice were supposed to be divided into 7 groups including model group （no treatment given）, sterile water group （intraperitoneal injection with sterile water）, treatment group （intraperitoneal injections with 1μg, 10μg, and 100μg doses of C19 polypeptid）, hydrocortisone group （ intraperitoneal injection with hydrocortisone） , and blank control group. After a 5- week-long course of medication started from the second day of modeling, a series of operations were necessary to be finished that including observing and scoring the skin lesions of mice, measuring the skinfold thickness, staining the samples of skin lesion with HE and toluidine blue, observing the pathology of skin lesions of mice and counting the mast cells. Results Under different concentrations, C19 polypeptide interventions give uneven alleviations to skin erythema, scales and edema of AD-like mice, and the same roles were played in decreasing the inflammatory cells infiltration. Conclusion Likely being related to inhibiting the immune reaction mediated by inflammatory cell, CKLF1-C19 is able to ease the inflammatory reaction of AD-like Murine Model.
