摘 要:目的:探讨人参皂苷-Rf对骨肉瘤细胞和人成纤维细胞是否具有细胞凋亡和DNA损伤的作用。方法:体外培养骨肉瘤细胞系(MG-63,U-2OS,HOS和SAOS-2)和人成纤维细胞;利用MTT法检测人参皂苷-Rf对骨肉瘤细胞系和人成纤维细胞增殖的影响;再用彗星电泳和γH2AX焦点实验检测人参皂苷-Rf诱导MG-63和U-2OS细胞的DNA损伤作用;最后利用DNA梯度实验观察人参皂苷-Rf对受损人成纤维细胞的修护作用。结果:MTT显示人参皂苷-Rf对骨肉瘤细胞增殖有抑制作用。人参皂苷-Rf诱导骨肉瘤MG-63、U-2OS细胞凋亡和DNA损伤,随浓度和处理时间增加而显著增加;进一步发现人参皂苷-Rf可修复和保护发生DNA损伤和凋亡(MNNG诱导)的人成纤维细胞。结论:人参皂苷-Rf可抑制骨肉瘤细胞的增殖,同时保护和修复受损的正常细胞。因此人参皂苷-Rf不仅可以作为抗癌药物使用,同时与其它抗癌药物配合使用可以有效的降低抗癌药物对正常细胞的副作用,保护正常细胞,使抗癌药物更好的发挥作用。Objective:To evaluate the potential genotoxicity of ginsenosides-Rf in human osteosarcoma cells(MG-63,U-2OS,HOS and SAOS-2)and the protective effect of ginsenosides-Rf on MNNGinduced DNA damage and apoptosis in human fibroblasts. Methods:The cytotoxicity of ginsenoside-Rf was determined by MTT assay. Employed alkaline comet assay,γH2AX foci formation were used to detect the DNA damage of MG-63 and U-2OS cells and the protection of ginsenoside-Rf against MNNG-induced genotoxicity in fibroblasts. Results:MG-63 and U-2OS cells were more sensitive to ginsenoside-Rf by MTT assay. As expected,ginsenosides-Rf induced significant concentration-dependent increases in DNA damage. The percentage of γH2AX positive reaction in MG-63 and U-2OS also revealed ginsenosides-Rf caused DNA double strand breaks(DSBs)in a concentration-dependent manner. Moreover,we detected that ginsenosides-Rf reduced MNNG-induced DNA damage and apoptosis in fibroblasts. Conclusion:Ginsenosides-Rf has obvious genotoxicity in human osteosarcoma cells. Situation instead,ginsenosideRf can protect against MNNG-induced genotoxicity in human normal cells. It suggested that ginsenosidesRf not only can kill cancer cells,but also serve as a natural cytoprotective agent against environmental carcinogens.
