摘  要:目的:建立随机案件海洛因滥用人员尿液的代谢组学研究模型,研究海洛因滥用与人体内生物标记物特异性变化关系在研究海洛因依赖的潜在价值。方法:随机选取日常案件中符合年龄、疾病史、性别比等要求,且经体金单克隆尿检板初筛、气质联用仪(GC/MS)确证滥用过海洛因的尿液30份,同时选取30例健康志愿者尿液,并用UPLC-MS分析尿液,经Mzmine 2.1软件峰匹配和多种统计学分析,筛选出有差异性变化的海洛因滥用生物标记物。再收集6份尿液重复实验对结果进行验证。结果:健康志愿者尿液与首次实验和验证实验的海洛因滥用者尿液可以实现分离,后两者存在交叉现象,说明健康组与滥用组尿液中生物标记物间存在特异性差异。结论:海洛因滥用形成依赖使得尿液中生物标记物发生了变化,经统计学初步筛选出90个特异性的生物标记物,说明借助代谢组学技术研究尿液中生物标记物的特异性变化来推断机体是否对海洛因依赖具有一定的可行性。Objective:To establish the model of metabonomics of urine in daily heroine abused cases, explore the potential value of the relationship between heroine abuse and biomarkers in urine of heroine addicts. Methods : The urine of heroin addicts in daily cases and health volunteers were collected, the endogenous metabolite in urine between two groups were compared and analyzed, the effect of heroin abuse on the metabolome in urine by UPLC - MS. We screened the differences in changes of heroine dependence biomarkers and repeated experiments were used to verify the results. Results:The data of urine in two groups have separation phenomenon which has been demonstrated the statistic differences. Conclusion:Heroin affected the metabolome in urine and the effect was specific and reproductive. Ninety potential biomarkers changed after initial screen, which establish the feasibility of Metabonomics in confirming the heroin dependence.
