摘  要:目的:探讨不同强度运动对青春期肥胖大鼠Vaspin表达的影响.方法:建模7周后,取32只高脂诱导的肥胖倾向大鼠,随机分为4组,每组8只,分别为安静对照组(OC)、低强度运动组(OL)、中强度运动组(OM)、高强度运动组(OH).OL、OM、OH分别以15-18m/min、21-25 m/min和28-32 m/min以及坡度均为0°的强度运动,1h/d,5次/周,干预8周.8周后采集麻醉后大鼠血液及内脏脂肪组织,用qRT-PCR测脂肪组织Vaspin mRNA基因表达,免疫组化测脂肪组织Vaspin蛋白表达,ELISA测血浆Vaspin浓度.结果:8周不同强度运动干预后,大鼠体重、内脏脂肪重量、体脂率较OC均下降明显(P<0.01),体重OL与OH差异显著(P<0.05),OL的内脏脂肪重量和体脂率较OM、OH均有差异(P<0.05、P<0.01);内脏脂肪组织Vaspin mRNA基因表达量OC与OH存有显著差异(P<0.05);OL、OM、OH的Vaspin蛋白表达较OC呈渐进式上升,强度越大,表达越明显;血浆Vaspin浓度OH较OC上调明显(P<0.05).结论:实施8周不同强度运动后,较好地降低了青春期肥胖大鼠体重、内脏脂肪重量和体脂率,运动强度越大,效果越显著.不同强度运动干预显著上调了Vaspin mRNA基因表达量、Vaspin蛋白表达及血浆Vaspin浓度,运动强度越大,上调幅度越明显.Objective: In order to investigate the effect ot vaspm aaolescem obesity exercise. Methods: High fat fed rat model after 7 weeks, take 32 obesity rats were randomly divided into 4 groups, 8 rats in each group, respectively, the control group(OC) , low intensity exercise group(OL), moderate intensity exercise group(OM), high intensity exercise group(OH). OL, OM, OH in animal treadmill running speed respectively for 15 - 18m/min, 21 - 25m/rain, 28 - 32m/rain, at an incline of 0 degrees, lh/day, 5 times/week, for 8 consecutive weeks. Anesthetized rats blood and visceral adipose tissue after 8 weeks, expression by qRT-PCR measurement of adipose tissue Vaspin mRNA, immunohistochemistry was used to measure the expression of Vaspin protein in fat cells, measurement of plasma Vaspin concentration by ELISA. Results: The 8 week exercise intervention, body weight, visceral fat weight and FAT% in adolescent rats were significantly lower than OC(P 〈 0.01) ;Body weight in OL and OH were different(P 〈 0.05 ) ;OM, OH rats, visceral fat weight and FAT% were lower than OL(P〈0.05,P〈0.01); In the Vaspin mRNA, the OC and OH groups were significant difference (P 〈0.05); The different intensity exercise prompted the expression Vaspin protein showed upward trend for OC, exercise intensity bigger and more obvious. In the concentration of plasma Vaspin, the level of OH was higher than OC(P 〈 0.05). Conclusion : 8 weeks of exercises of different intensity significantly reduce body weight, visceral fat weight and FAT%, exercise intensity bigger and more obvious. Three different intensity exercise prompted the expression Vaspin mRNA, Vaspin protein in visceral adipose tissue of rat and the level of plasma Vaspin to show upward trend, exercise intensity bigger and more obvious.
