Influence of Xuebijing Injection on Formation of Angiopoietin 2 and Cadherin in Vascular Endothelial Cells Injured by Traumatic Stress

摘  要:【目的】观察细胞外热休克蛋白70(e HSP70)模拟创伤刺激内皮细胞后血管生成素2(Ang-2)及血管内皮钙黏蛋白(VEcadherin)的变化,探讨血必净注射液对血管内皮的保护作用。【方法】培养人脐静脉内皮细胞,检测e HSP70刺激0、2、4、8、16、32 h后细胞Ang-2、VE-cadherin的m RNA和蛋白表达量,找出最佳刺激时间,然后设空白对照组、e HSP70组(终浓度为0.1μg/m L)、血必净组(终浓度为50 mg/m L)、血必净+e HSP70组共4个实验组,检测最佳时间点各组上述指标改变。【结果】e HSP70刺激后Ang-2的m RNA及蛋白表达量增加并于4 h达到高峰,而VE-cadherin的m RNA和蛋白表达量经e HSP70刺激后降低,并于4 h达到最低值。各实验组干预4 h后,e HSP70组与血必净+e HSP70组细胞Ang-2的m RNA及蛋白表达量较空白对照组显著增高(P<0.05),而血必净+e HSP70组的表达量较e HSP70组显著下降(P<0.05);与空白对照组相比较,e HSP70组VE-cadherin的m RNA及蛋白的表达量显著降低(P<0.05),而血必净+e HSP70组及血必净组上述指标表达量均显著上升(P<0.05),其中尤以血必净组的表达增高最为显著(P<0.05)。【结论】血必净注射液通过调控e HSP70对Ang-2与VE-cadherin的表达影响,对创伤刺激的血管内皮细胞具有一定的保护作用。Objective To observe the changes of angiopoietin 2 (Ang-2) and vascular endothelial cadherin (VE- cadherin) in vascular endothelial cells injured by analogue trauma stress induced by intracellular heat shock protein 70 (eHSP70), and to explore the protective effect of Xuebijing Injection (XI) on the endothelial cells. Methods The expression levels of Ang-2 and VE-cadherin mRNA and protein in the cultured human umbilical vein endothelial cells(HUVEC)were monitored at hour 0, 2, 4, 8, 16, 32 after eHSP70 stimulation for the screening of the optimal stimulating time. The cultured HUVEC were randomized into blank control group, eHSP70 group(eHSP70 at final concentration of 0.1 μg/mL), XI group(XI at final concentration of 50 mg/mL), and XI + eHSP70 group, and the levels of Ang-2 and VE-cadherin at the optimal stimulating time were observed. Results After eHSP70 stimulation, the quantity of Ang-2 mRNA and protein expression in HUVEC was increased and then arrived at the peak value at hour 4, and the quantity of VE-cadherin mRNA and protein expression was decreased and then arrived at the valley value at hour 4. After medicine intervention for 4 h, the quantity of Ang-2 mRNA and protein expression in HUVEC of eHSP70 group and XI +eHSP70 group was increased (P〈0.05 compared with that in the blank control group) , and XI+eHSP70 group had lower Ang-2 mRNA and protein level than eHSP70 group (P〈0.05). Compared with the blank control group, the quantity of VE-cadherin mRNA and protein expression in eHSP70 group was decreased obviously (P〈0.05) , and XI+ eHSP70 group and XI group had higher VE-cadherin mRNA and protein level than eHSP70 group (P〈0.05), in particular XI group had the highest level (P〈0.05). Conclusion XI has protective effect on traumatic-stress- induced endothelial cells injury through regulating Ang-2 and VE-cadherin expression medicated by eHSP70.
