摘 要:目的观察姜黄素对泡状棘球蚴原头节生长的影响。方法将体外培养的泡状棘球蚴原头节分为6组:空白对照组,溶剂对照组和姜黄素20、40、60、80μmol/L组,观测原头节活性并计算存活率;姜黄素作用原头节24h后,应用半胱天冬氨酸蛋白酶-3(caspase-3)检测试剂盒检测caspase-3活性,应用原位末端脱氧核糖核苷酸转移酶标记技术(TUNEL法)试剂盒检测原头节细胞凋亡情况。结果培养9d后,空白对照组与溶剂对照组原头节活力无明显改变,各浓度姜黄素干预组原头节存活率与对照组相比差异均有统计学意义(F80μmol/L=4.986,F60μmol/L=4.826,F40μmol/L=4.364,F20μmol/L=3.288,P<0.05)。姜黄素对原头节的抑制作用呈时间、浓度依赖,80μmol/L姜黄素作用9d时原头节全部死亡。不同浓度姜黄素组作用24h原头节,caspase-3活性与对照组相比显著增高(F=446.937,P<0.05)。TUNEL法检测姜黄素干预原头节阳性率显著高于对照组(χ2=7.977,P<0.05)。结论姜黄素对体外培养的原头节有一定杀灭作用且具有时间和浓度依赖性。姜黄素对头节的杀灭作用可能与诱导头节细胞凋亡有关,其机制尚有待进一步研究。Objective To research the in vitro effects of curcumin on Echinococcus multilocularis protoscoleces.Methods E.multilocularis protoscoleces were cultured in vitroin a blank control group,in the presence of a solvent,or in the presence of curcumin at a concentration of 20,40,60,or 80μmol/L.Protoscoleces were subjected to 0.1% eosin staining and observed under an inverted microscope to determine their survival rate and motility.Caspase-3activity was assessed with a caspase-3assay kit.The apoptosis of protoscoleces was determined with a TdT-mediated dUTP Nick-End Labeling assay. Results There were no marked changes in the survival of E.multilocularis protoscoleces in the blank control group and the group treated with a solvent.There were significant differences in the survival rate of E.multilocularis protoscoleces in the control group and groups treated with different concentrations of curcumin(F80μmol/L=4.986,F60μmol/L=4.826,F40μmol/L=4.364,F20μmol/L=3.288,P〈0.05).After 9d,there were no surviving protoscoleces in the group treated with curcumin at a concentration of 80μmol/L.Curcumin inhibited protoscoleces in a time-and concentration-dependent manner.The results of caspase-3detection indicated that caspase-3activity was significantly higher in protoscoleces after 24 hof incubation in curcumin(F=446.937,P〈0.05).Apoptosis in the group treated with curcumin at a concentration of 80μmol/L was mainly " +," with an apoptosis rate of 31.6%,and " ++," with an apoptosis rate of28.9%;these rates were significantly higher than that in the control group according to a TUNEL assay(χ2=7.977,P〈0.05). Conclusion Curcumin inhibited E.multilocularis in vitroin a time-and concentration-dependent manner.Its mechanism of action may involve its induction of apoptosis.However,further studies on the specific mechanisms of action of curcumin were needed.
