摘 要:目的对一起感染性腹泻疫情的未知种型气单胞菌进行病原学鉴定。方法采集本起疫情腹泻患儿粪便标本用碱胨水增菌后接种于麦康凯平板,对可疑菌落进行系统生化鉴定;对鉴定为气单胞菌的菌株提取核酸,进行rpoB基因扩增及测序分析,所得序列在GenBank中进行同源性搜索并构建分子系统树,鉴定致感染性腹泻的气单胞菌基因种。结果 6份腹泻患儿粪便标本经培养后分离鉴定出4株气单胞菌,对rpoB基因测序后进行同源性搜索及系统发生分析,分离株气单胞菌为豚鼠气单胞菌。结论引起本次感染性腹泻疫情的病原菌为豚鼠气单胞菌。Objective To identify the unknown species of Aeromonas that caused an outbreak of infectious diarrhea.Methods Fecal specimens were collected from patients with diarrhea and enriched in APW.Specimens were then plated on MAC plates.Suspected bacteria were identified using conventional biochemical methods.The DNA of Aeromonas strains was extracted.The rpoB gene was amplified and sequenced.A sequence homology search was conducted on GenBank,and a molecular phylogenetic tree was constructed to determine the genotype that the Aeromonas strain belonged to. Results Four Aeromonas strains were isolated from 6specimens using conventional techniques.The RpoB gene of the 4 Aeromonasisolates was sequenced and analyzed.Results indicated that interspecies divergence of the 4 Aeromonasisolates and A.caviae was 0.00-0.13.The 4 Aeromonasisolates and A.caviae ATCC15468 were clustered in the same group.The species most closely related to the isolates was A.caviae.The sequence homology search and molecular phylogenetic tree indicated that the Aeromonasisolate was A.caviae. Conclusion The pathogen that caused an outbreak of infectious diarrhea was A.caviae.
