17味中药对豚鼠皮肤TNF-α、IL-10作用的影响

摘　　要：目的探讨17味中药对豚鼠皮肤肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)的调节作用。方法实验动物随机分4组,17味中药乙醇提取液为实验组,8-甲氧补骨脂素(8-MOP)、75%乙醇为阳性及阴性对照组,均涂于豚鼠皮肤上,40 d后取皮肤标本免疫组化染色,测TNF-α、IL-10平均光密度,比较其作用结果。结果当归、甘草、枸杞、红花、女贞子、首乌、益母草、黑芝麻、栀子对TNF-α表达下调;甘草、枸杞、合欢皮、黄芪、女贞子、沙参、沙苑子、菟丝子、黑芝麻对IL-10表达上升,其中甘草、枸杞、女贞子、黑芝麻对TNF-α、IL-10均表现正向调节作用;合欢皮、黑桑葚对TNF-α表达上调,旱莲草、黑桑葚使IL-10表达下降,出现负向调节作用。结论通过中药对TNF-α、IL-10调节作用探讨,为研制治疗白癜风的有效组方提供可靠实验数据。Objective In order to study effects of the 17 kinds of traditional Chinese medicine （TCM） on tumor necrosis factor-α （TNF-α）, Interleukin-10 （IL-10） in guinea pig skin. Methods The rats were divided into four groups randomly, 17 kinds of herbal extract in alcohol for the experimental group, 8-methoxypsoralen （8-MOP） and 75% ethanol for positive group and negative group, coated on the skin of guinea pigs, forty days later cut off sample skin and process it with immunohistochemical staining,measured the mean optical density of TNF-α and IL-10, and compared the results. Results The expression of TNF-ct was decreased in angelica, liquorice, wolfberry, safflower, ligustrum lucidum ait, polygonum multiflorum, herba leonuri, black sesame and gardenia. The expression of IL-10 was increased in liquorice, wolfberry, albizia, astragals, Ligustrum lueidum air, adenophora, eomplanatus, semen cuseutae and black sesame. Among them, liquorice, wolfberry, ligustrum lucidum ait and black sesame had a positive effect on the expression of TNF-α and IL-10. The high expression of TNF-α in albizia and black mulberry and the low expression of IL-10 in eelipta and black mulberry had negative effects. Conclusion The Chinese medicine regulating effect on TNF-α and IL-10 study provide reliable experimental data for the development of effective prescription for the treatment of vitiligo.
