摘 要:目的探讨舒郁胶囊治疗经前期综合征(PMS)肝气郁证的可能作用机制。方法将60只Wistar大鼠随机分为空白组、模型组、舒郁胶囊组、氟西汀组、柴胡皂苷组,每组12只。除空白组外,各组大鼠均于动情周期行四肢对侧捆缚术造模。造模同时,舒郁胶囊组给予舒郁胶囊0.408 g/(kg·d)、氟西汀组给予盐酸氟西汀胶囊2.67 mg/(kg·d)、柴胡皂苷组给予柴胡皂苷提取物0.72 mg/(kg·d),空白组与模型组给予等体积无菌水灌胃,连续灌胃8天。给药后称量大鼠体重,进行旷场实验采集旷场实验总路程,并取大鼠顶区皮层组织检测γ-氨基丁酸转氨酶(GABA-T)、γ-氨基丁酸转运体-1(GAT-1)蛋白和mRNA表达情况。结果与空白组相比,模型组大鼠体重下降、旷场实验总路程减少(P<0.05),顶区皮层GABA-T、GAT-1蛋白和mRNA表达显著升高(P<0.05);与模型组相比,氟西汀组、舒郁胶囊组大鼠体重增长、旷场实验总路程增长(P<0.05),顶区皮层GABA-T、GAT-1蛋白及mRNA的表达显著下降(P<0.05)。结论舒郁胶囊可能通过纠正GABA-T、GAT-1的异常表达调节GABA代谢失常而发挥治疗PMS肝气郁证的作用。Objective To discuss possible mechanism of Shuyu Capsule (舒郁胶囊 ) treating premenstrual syndrome with liver-Qi depression syndrome. Methods Sixty Wistar rats were randomly divided into blank group, model group, Shuyu Capsule group, the fluoxetine group and the saikoside group, 12 rats in each group. Except for the blank group, contralateral limbs of rats in other group were bound to make model in estrous cycle. At the same time, the Shuyu Capsule group was given Shuyu Capsule 0. 408 g/( kg · d). The fluoxetine group was given fluoxetine hydrochloride capsule 2.67 mg/( kg · d). The saikoside group was given extractive of saikoside 0.72 mg/( kg· d). The blank group and the model group were given the isopyknic sterile water to garage for 8 days. The researchers weighed those rats after medication. The rats were made open field test to collect the total distance. And the researchers extracted cortex tissue of parietal region of rats to detect expression of γ-aminobutyric acid transaminase ( GABA-T), γ-aminobutyric acid transporter-1 (GAT-1) protein and mRNA. Results Compared with the blank group, weight of rats and the total distance of open field test reduced in the model group ( P 〈 0.05 ), but expression of GABA-T, GAT-1 protein and mRNA in the cortex of parietal region significantly increased ( P 〈 0. 05 ). Compared with the model group, body weight and the total distance of open field test increased in the fluoxetine group and the Shuyu Capsule group (P 〈0.05), but expression of GABA-T, GAT-1 protein and mRNA in the cortex of parietal region significantly decreased (P 〈 0. 05 ). Conclusion Shuyu Capsule may play therapeutic role to premenstrual syndrome with liver-Qi depression syndrome by correcting the abnormal expression of GABA-T, GAT-1 to regulate disordered metabolism of GABA.
