摘  要:目的:探讨茵芍散含药血清对大鼠肝星状细胞(HSC)增殖作用的影响及其机制。方法:40只Wistar大鼠分别给予中药高、中、低剂量、0.9%氯化钠溶液、己酮可可碱灌胃,喂养1周后,腹主动脉采血,制备药物血清。设中药高、中、低剂量组,阴性对照组,阳性对照组及空白组,药物血清干预HSC-T6后,运用流式细胞术、吖啶橙/溴化乙锭(AO-EB)染色观察细胞增殖、分析周期情况,并分别通过实时荧光定量PCR、Western blot检测β-catenin m RNA及蛋白表达情况。结果:与对照组比较,茵芍散能有效抑制HSC-T6增殖,下调β-catenin m RNA及蛋白表达(P<0.05),且具有量-效关系。结论:茵芍散可抑制HSC增殖,阻断Wnt/β-catenin信号通路对HSC的激活可能是其治疗肝纤维化的作用机制。Objective: To explore the influence and mechanism of serum containing Yinshao Powder on proliferation of hepatic stellate cells(HSC) of rats. Methods: Forty Wistar rats received gavage administration of low, middle and high dosage of traditional Chinese medicine(TCM), normal saline(NS), and pentoxifylibe(PTX) respectively for a week, then, the abdominal blood samples were obtained for the preparation of medicated serum. HSC-T6 cells in control group, PTX group, blank group,low, middle and high dosage of serum containing Yinshao Powder group were intervened with corresponding drugs, and then the proliferation and cell cycles of cells were observed by using flow cytometry(FCM) and acridine orange-ethidium bromide(AOEB) staining method. The m RNA and protein expression of β-catenin were tested by using PCR and Western blot respectively. Results: Yinshao Powder could control the proliferation of HSC-T6, and decrease the expression of β-catenin m RNA and protein, and the difference was statistically significant compared with the control group(P〈0.05). Conclusion: Yinshao Powder could inhibit the proliferation of HSC, and block the signaling pathway of Wnt/β-catenin, which might be its curative mechanism on hepatic fibrosis.
