摘 要:目的:观察黄芪有效组分对脂多糖诱导的大鼠肾小球系膜细胞表达血小板源性生长因子-BB(PDGFBB)的影响。方法:将黄芪有效组分(黄芪多糖、黄芪甲苷、毛蕊异黄酮)作用于经脂多糖刺激的大鼠肾小球系膜细胞(GMCs)后,采用逆转录-聚合酶链反应法、免疫细胞化学法检测PDGF-BB m RNA、蛋白的表达。结果:模型组经脂多糖刺激后PDGF-BB m RNA及蛋白表达增加(P<0.05),黄芪不同组分均能不同程度抑制PDGFBB m RNA及蛋白表达(P<0.05)。结论:说明黄芪多糖、黄芪甲苷、毛蕊异黄酮可不同程度抑制GMCs PDGF-BB m RNA及蛋白的表达,从而改善或延缓系膜增生性肾病肾小球的硬化。Objective: To observe the effects of effective components in astragalus on the lipopolysaccharides(LPS)-induced platelet-derived growth factor-BB(PDGF-BB) expression of glomerular mesangial cells(GMCs) in rats. Methods: Protein and m RNA expression of PDGF-BB were respectively detected by immunocytochemistry and reverse transcriptase-polymerase chain reaction after effective components in astragalus(astragalus polysaccharide, astragaloside IV and calycosin) acting on LPSinduced GMCs. Results: The protein and m RNA expression of PDGF-BB in GMCs were enhanced(P〈0.05) after the stimulation of LPS, and astragalus components significantly inhibited the m RNA and the protein expression of PDGF-BB at different levels(P〈0.05). Conclusion: Astragalus polysaccharide, astragaloside IV and calycosin could inhibit the m RNA and protein expression of PDGF-BB, which could further improve and slow down the sclerosis of glomerulus caused by mesangial proliferative glomerulone phropathy.
