摘  要:目的:研究川芎嗪对流感病毒H1N1感染A549细胞诱导的凋亡干预作用,探讨川芎嗪抗流感病毒感染的作用机制。方法:采用荧光定量PCR检测各组细胞中天门冬氨酸特异性半胱氨酸蛋白Caspase-3、Caspase-8和Caspase-9 m RNA表达的变化。结果:与细胞对照组比较,H1N1感染组Caspase-3、Caspase-8和Caspase-9 m RNA表达均明显升高(P<0.01)。与H1N1感染组比较,奥司他韦对照组及川芎嗪高、低剂量组的Caspase-3、Caspase-8、Caspase-9 m RNA表达均明显降低(P<0.01,P<0.05)。结论:流感病毒体外感染细胞A549后诱导凋亡,川芎嗪能通过调控天门冬氨酸特异性半胱氨酸蛋白家族介导凋亡通路的相关基因表达,对病毒感染诱导的细胞凋亡有干预作用,从而发挥抗病毒作用。Objective: To investigate the intervention effect and mechanism of ligustrazine on apoptosis of A549 cells induced by virus H1N1. Methods: The m RNA expressions of Caspase-3, Caspase-8, and Caspase-9 were verified by using q PCR respectively. Results: The results of q PCR showed that the expression of m RNA of Caspase-3, Caspase-8, and caspase-9 in H1N1 infection group was higher than that of control group significantly(P〈0.01). Compared with H1N1 infection group, the epression of m RNA of Caspase-3, Caspase-8, and Caspase-9 in Oseltamivir group, low and high dosage of ligustrazine groups were decreased significantly(P〈0.01, P〈0.05). Conclusion: In the apoptosis of A549 cells induced by H1N1 virus in vitro, ligustrazine could intervene the apoptosis process induced by virus through regulating the expression of relative genes in caspase protein family mediated apoptosis pathway.
