摘  要:目的探讨聚丙烯酸树脂包裹白介素21DNA核酸疫苗免疫Babl/c小鼠的作用,检测小鼠CTL细胞抗daudi细胞的作用。方法采用乳剂—溶剂挥发法制备微球疫苗,分组免疫BALB/c小鼠,将制备所得的抗Daudi细胞肿瘤特异性CTL细胞作用于Daudi细胞,通过MTT法计算Daudi细胞增值抑制率,流式细胞仪测量Daudi细胞凋亡率,检测CTL细胞的杀伤作用。结果光学显微镜观察微球疫苗粒径约为(50.6±12.2)μm,且微球疫苗组对Daudi细胞的杀伤作用明显高于核酸疫苗单独免疫组。结论聚丙烯酸树脂能有效地保护核酸疫苗,并能提高其在细胞内的表达,为肿瘤疫苗的后续研究提供了思路和理论依据。Objective To explore the functions of polyacrylic acid resin package of IL21 DNA nucleic acid vaccine to immunize Babl/c mice.The reaction of CTL cells to anti-Daudi cells was detected.Methods The microsphere vaccines were prepared with an emulsion solvent evaporation method.The CTL cells against Daudi cell were prepared by immunized BALB/c mouse.The rate of cell proliferation of Daudi cells was calculated,and the flow cytometry was used to measure the apoptosis rate.Results The particle size of microsphere vaccine was approximately 550.6±12.2μm observed by the optical microscope.The proliferation rate and apoptosis rate of Daudi cells were significantly higher than that of nucleic vaccine groups alone.Conclusion Polypropylene acid resin can effectively protect the nucleic acid vaccine,and can improve its expression in the cells.The results of this study provide a new theoretical basis for the following study in tumor vaccine.
