六味地黄颗粒对哮喘大鼠促上皮生长因子及其受体干预作用的研究

摘  要:目的探讨六味地黄颗粒对哮喘大鼠气道重塑过程中促上皮生长因子(EGF)及促上皮生长因子受体(EGFR)可能的干预作用。方法用ELISA法检测哮喘大鼠肺泡灌洗液中的EGF及EGFR水平,比较空白对照组、哮喘模型对照组、各用药组之间两种蛋白水平的差异。结果哮喘模型对照组的肺泡灌洗液EGFR水平较其他各组明显增高(P<0.01);六味地黄颗粒+布地奈德组、空白对照组及布地奈德组之间EGFR水平差异无统计学意义(P>0.05);六味地黄颗粒治疗组的肺泡灌洗液中EGFR水平居于哮喘模型对照组和空白对照组之间。哮喘模型对照组EGF水平最高,明显高于其他各组(P<0.01);空白对照组与六味地黄颗粒+布地奈德组EGF水平差异不大(P>0.05),但均显著低于其他三组(P<0.01);六味地黄颗粒治疗组EGF水平明显高于布地奈德组,差异有统计学意义(P<0.01)。结论六味地黄颗粒能降低哮喘大鼠肺部促上皮生长因子及其受体的生成,但其降低作用不如布地奈德明显。Objective To investigate possible intervention effects of Liuwei Dihuang granule on epidermal growth factor (EGF)and epidermal growth factor receptor(EGFR)of asthma model rats.Methods EGF and EGFR levels in bronchoalve-olar lavage fluid of asthmatic rats were detected by ELISA method.And then,difference of the two protein levels between blank control group,asthma model control group and each treatment group were compared.Results EGFR levels in bronchoalveo-lar lavage fluid of asthma model control group were significantly higher than those of the other groups(P〈0.01).Difference of EGFR levels in bronchoalveolar lavage fluid of Liuwei Dihuang granule+budesonide group,blank control group and budes-onide group was not statistically significant(P〉0.05).EGFR levels in bronchoalveolar lavage fluid of the Liuwei Dihuang granule group was between those of the asthma model control group and blank control group.Asthma model control group had the highest EGF level,significantly higher than those of other groups(P〈0.01).No significant difference was found in EGF levels of the Liuwei Dihuang granule+budesonide group and blank control group(P〉0.05),but they were all obviously lower than the three other groups(P〈0.01).In addition,EGF levels of the Liuwei Dihuang granule group were obviously higher than those of the budesonide group,so difference was statistically significant(P〈0.01).Conclusion Traditional Chinese medicine Liuwei Dihuang granule can reduce the EGF and EGFR levels of asthma model rats,but its reducing effect is less obvious than budesonide.
