冬病夏治中药敷贴对哮喘大鼠树突状细胞免疫功能的影响

摘　　要：目的:验证"冬病夏治"穴位敷贴疗法能通过下调树突状细胞的表达,抑制哮喘的变应性炎症,增强树突状细胞的免疫功能,从而减少哮喘发作的设想。方法:以卵清蛋白(VOA)致敏大鼠并诱发哮喘模型,用ELISA方法检测血清中OVA特异的Ig E水平,比较穴位敷贴组,哮喘模型组,正常对照组之间有无差异;同用此法检测气管肺泡灌洗液(BALF)中IL-4、IL-5水平,比较各组之间有无差异。通过大鼠肺脏DC稳定的培养方法,用流式细胞仪检测肺脏DC表面分子CD11c CD80、CD11c CD86的表达,比较各组树突状细胞表面分子表达水平有无差异。结果:与健康对照组相比,穴位敷贴组血清中VOA特异的Ig E水平无差异(P>0.05),BALF中IL-4、IL-5水平无差异(P>0.05),树突状细胞表面分子表达水平无差异(P>0.05)。与哮喘模型组相比,穴位敷贴组VOA特异的Ig E水平有差异(P<0.05),BALF中IL-4、IL-5水平有差异(P<0.05),树突状细胞表面分子表达水平有差异(P<0.05)。结论:穴位敷贴疗法能通过下调树突状细胞的表达,抑制哮喘变应性炎症,从而减少哮喘的发作。Objective： Verifying that ＂ winter disease in summer treat＂ acupoint therapy could by reduced expression of dendritic cells,inhibiting the allergic inflammation of asthma,enhanced the immune function of dendritic cells,thereby reducing the idea of asthma attacks. Methods： Egg white protein（ VOA） and asthma in sensitized rats model with ELISA method for detection of serum OVA Ig E levels,comparing the differences among acupoint group, asthma model group, and controlled group; with trachea was examined by the bronchoalveolar lavage fluid（ BALF） levels of IL-4,IL-5,there was no difference between the groups. Through the DC stable cultivation method of lungs in rats by flow cytometry lung DC surface molecules expression of CD11 c CD80,CD11 c CD86,dendritic cell surface molecules expression level in each group there was no difference.Results： Compared with the controlled group,model dendritic cell surface molecules expression level（ P〈0. 05）,compared with the model group,acupoint group decreased dendritic cell surface molecules expression level（ P〈0. 05）. Conclusion： Acupoint therapy can reduce asthma attacks by reducing expression of dendritic cells and suppressing asthma allergic inflammation.
