摘  要:目的探讨瓜蒌皮总黄酮对低密度脂蛋白(LDL)诱导的血管内皮损伤的保护作用,为瓜蒌皮有效成分筛选提供新思路。方法将40只SD大鼠随机分为空白组、模型组、瓜蒌皮总黄酮高剂量组、瓜蒌皮总黄酮中剂量组和瓜蒌皮总黄酮低剂量组,每组8只。空白组给予生理盐水,模型组给予血清LDL(4 mg/kg)诱发血管内皮功能损伤,瓜蒌皮总黄酮高、中、低剂量组,用不同剂量总黄酮(90、30、10 mg/kg)每天灌胃1次,连续5 d,药物处理第4天舌下静脉注射LDL(4 mg/kg)。实验结束后检测肿瘤坏死因子-α(TNF-α)、非对称性二甲基精氨酸(ADMA)、丙二醛(MDA)和血浆中一氧化氮(NO)的浓度,并取大鼠胸主动脉环做血管舒张实验。结果与空白组比较,模型组大鼠血清TNF-α、ADMA和MDA升高,NO浓度降低(P<0.01);高、中剂量瓜蒌皮总黄酮能抑制LDL所致血清TNF-α、ADMA和MDA含量的升高,NO浓度的降低(P<0.05);低剂量瓜蒌皮总黄酮仅抑制MDA含量的升高(P<0.05);血管舒张实验结果显示,瓜蒌皮总黄酮能显著改善大鼠胸主动脉环舒张反应,并且呈剂量依赖性,剂量越大,大鼠胸主动脉环舒张反应越强。结论瓜蒌皮总黄酮对血管内皮损伤具有保护作用。Objective To probe the protective effect of trichosanthes skin flavonoids( TSF) on damages of the endothelium induced by Low- density lipoprotein( LDL),and provide new ideas for the active ingredient screening of Trichosanthes skin. Methods 40 SD rats were randomly divided into five groups( blank group,model group,TSF high dosage group,TSF moderate dosage group,TSF low dosage group) with each group 8 rats,the blank group treated with normal saline,the model treated with the injection of native LDL( 4 mg /kg) in rats to induce vascular endothelial injury,TSF high dosage group,TSF moderate dosage group,TSF low dosage group treated with 90,30,10 mg / kg flavonoids gavage one time a day repectively for5 days and LDL( 4 mg / kg) vein under ligua injection on the the fourth day. Blood samples were collected from carotid artery,serum concentrations of tumour necrosis factor- α( TNF- α),nitric oxide( NO),asymmetric dimethylarginine( ADMA) and malondialdehyde( MDA) were measured,and vasodilation experiments with rat thoracic aorta rings were performed. Results Contrasted with the blank group,the model group increased in the levels of TNF- α,ADMA and MDA,and decreased in the serum levels of NO significantly( P〈 0. 01); TSF low dosage group inhibit the increase of the content of MDA significantly( P 〈 0. 05),the results of vasolidation experiment indicated that TSF could improve the vasolidator responses of thoracic aorta significantly with the greater the dose,the greater the vasolidator responses of thoracic aorta in rats. Conclusion Trichosanthes skin flavonoids has the function of protecting the damages of endothelium in rats.
