摘  要:目的探讨脂多糖(LPS)诱导星形胶质细胞产生炎性反应及姜黄素对它的抑制作用。方法在2015年3月—2015年7月期间,分离培养了10只新生1~2 d的昆明小鼠大脑皮质星形胶质细胞,培养10 d后传代分组,实验分PBS对照组(n=6)、LPS刺激组(n=6)、LPS联合姜黄素干预组(n=6)。ELISA检测炎性因子IL-6、IL-1β和TNF-α的释放。结果与PBS组的IL-6(873.82±20.28)、IL-1β(680.62±10.35)和TNF-α(492.73±8.56)比较,LPS刺激组IL-6(1132.42±78.09)、IL-1β(821.87±14.33)和TNF-α(559.32±9.6)的分泌增加(P〈0.05);姜黄素能显著抑制LPS诱导的IL-6(1014.79±52.08)、IL-1β(738.84±7.33)和TNF-α(476.7±10.38)的分泌,与LPS组相比差异有统计学意义(P〈0.05)。结论姜黄素能抑制LPS激活星形胶质细胞所致的炎性反应。Objective To investigate the inflammatory response in astrocytes induced by lipopolysaccharide(LPS) and the inhibition of curcumin on inflammatory response. Methods And the PBS group IL-6(873.82 ± 20.28) IL-1 beta(680.62 ±of 10.35) TNF-a and(492.73 ± 8.56), LPS stimulated IL-6 group(1132.42 ± 78.09), IL-1 beta(821.87 ±14.33), and TNF-(559.32 ± 9.6) secretion increased(P〈0.05); curcumin could significantly inhibit the LPS induced IL-6(1014.79 ±52.08),interleukin 1 IL-1(738.84 ± 7.33) and tumor necrosis factor alpha(476.7 ± 10.38) secretion, and LPS group compared to the difference was statistically significant(P〈0.05). Results Compared with IL-6(873.82±20.28)、IL-1β(680.62±10.35)and TNF-α(559.32±9.6) of PBS control group,LPS significantly increased the production of IL-6(1132.42±78.09)、IL-1β(821.87±14.33) and TNF-α(559.32±9.6)(P〈0.05). Cur treatment inhibited LPS-induced IL-6(1014.79±52.08)、IL-1β(738.84±7.33) and TNF-α(476.7±10.38) production compared with LPS stimulated group, the differences were statistically significant(P〈0.05). Conclusion Cur could inhibit LPS-induced inflammatory response in astrocytes.
