摘 要:目的:优选五味子治疗失眠有效部位的提取工艺。方法:采用单因素试验考察回流提取时间、乙醇体积分数、溶剂用量、提取温度、提取次数等对五味子总木脂素含量的影响。在单因素试验基础上选取乙醇体积分数、乙醇用量、提取时间为考察因素,每个因素设置5个水平,以总木脂素得率为评价指标,利用星点设计-效应面法优选回流提取工艺。结果:五味子治愈失眠有效部位的最佳回流提取工艺为乙醇体积85%、乙醇用量3倍、提取时间49 min、提取次数1次。结论:优选的回流提取工艺简便易行,预测性良好,为五味子后续的剂型开发和工业化生产提供实验依据。Objective: To optimize extraction process of effective parts of Wuweizi for curing insomnia. Method: Single- factor tests were adopted to investigate effects of extraction time, ethanol concentration, solvent dosage, extraction temperature and extraction times on contents of total lignans in Wuweizi. Based on single factor tests, ethanol concentration, solvent dosage and extraction time were used as factors (five levels for each factor), then extraction process of effective parts in Wuweizi for curing insomnia was optimized by central composite design-response surface methodology with extraction efficiency of total lignans as evaluation indicator. Result: Optimum extraction process was as follows: extracted 49 rain with 3 volumes of 85% ethanol at 90℃, 1 time. Conclusion: Optimized extraction process is simple and feasible with good predictability. It can provide experimental evidence for formulations development and industrial production of Wuweizi.
