摘 要:目的:考察梓醇对脑缺血大鼠髓鞘的保护作用。方法:采用大鼠大脑中动脉缺血模型,将造模大鼠分成模型组、梓醇15mg/kg、30mg/kg、60mg/kg组、丁苯酞70mg/kg组和依达拉奉7mg/kg组和假手术组,缺血后3d至14d给药。采用Longa法对大鼠神经功能进行评分;HE染色和卢卡斯快蓝(luxolfastblue,LFB)染色观察胼胝体区髓鞘改变;免疫组织化学检测胼胝体髓鞘碱性蛋白(myelinbasicprotein,MBP)表达。结果:与假手术组比较,模型组大鼠的神经功能明显损伤;梓醇30mg/kg、60mg/kg组大鼠神经功能行为学评分明显降低。HE染色,模型组可见胼胝体白质疏松、空泡形成、胶质细胞深染、间质水肿;梓醇组胼胝体白质少许空泡形成、胶质细胞形态趋于正常。LFB染色,模型组髓鞘颜色变浅,髓鞘变得稀疏;梓醇30mg/kg、60mg/kg组积分光密度值显著升高。MBP蛋白在假手术组的表达较强,为黄褐色;在模型组表达明显减弱,着色较浅,且分布不均匀;梓醇30mg/kg、60mg/kg组MBP积分光密度值显著升高。结论:梓醇可促进大鼠脑缺血损伤后神经功能障碍的恢复,可能与髓鞘的保护作用有关。Objective: To investigate the effects of catalpol on protective myelin sheath in cerebral ischemia rats. Methods: The rat model was adopted by permanent middle cerebral artery ischemia. The model rats were randomly divided into six groups,model group,15 mg / kg,30 mg / kg,60 mg / kg group of catalpol,butylphthalide 70 mg / kg group,edaravone7 mg / kg group,sham-operated group,the rats were given medicine for 3 to14 days. Neural functional behavior score of rats were tested by Longa method. Myelin of orpus callosum were observed by HE staining and Lucas fast blue staining Myelin basic protein( MBP) expression in orpus callosum were detected by chemical colorimetry. Results: Nerve function of model group rats were obviously damaged; Neural functional behavior score of 30 mg / kg,60 mg / kg of catalpol group decreased obviously,HE staining,the corpus callosum white matter of model group is loose; glial cells vacuolation and hyperchromatic,interstitial edema; corpus callosum white matter of catalpol group has a little vacuolation,glial cells form returns to normal. LFB staining,color becomes shallow,myelin becomes spars of model group. 30 mg / kg,60 mg / kg of catalpol group integral optical density value increased significantly. MBP protein expression of control group is strong,for tan; Expression decreased significantly in the model group,coloring,and uneven distribution MBP integral optical density were significantly increased 30 mg / kg,60 mg / kg of catalpol group. Conclusion: Catalpol can promote the recovery of nerve dysfunction of ischemia injury rats,may be associated with protection of myelin.
