两头尖醇提物抗佐剂性关节炎作用的研究

摘　　要：目的:探索两头尖醇提物对佐剂性关节炎大鼠(AA)的抗炎作用。方法:取48只SD大鼠随机分为正常组、模型组、甲氨蝶呤片(3mg/kg)组和两头尖醇提物189mg/kg、378mg/kg、756mg/kg组,除正常组外其余各组大鼠右后足跖皮内注射弗氏完全佐剂(Frond's completed Adjuvant,CFA),诱发大鼠AA模型,记录大鼠体重,检测并计算足跖肿胀度、关节炎指数、胸腺指数和脾指数,ELISA法测定各组大鼠血清中IL-1、IL-6、IL-8、IL-10和TNF-α的含量。结果:与模型组相比,两头尖醇提物189mg/kg、378mg/kg、756mg/kg能增加体重,能抑制AA大鼠原发性足肿胀,降低继发性AA大鼠关节炎指数、胸腺指数和脾指数,此外还能下调促炎性细胞因子IL-1、IL-6、IL-8、TNF-α水平,上调抑炎性细胞因子IL-10的水平。结论:两头尖醇提物189mg/kg、378mg/kg、756mg/kg组对佐剂性关节炎大鼠原发性和继发性炎症均有显著的治疗作用。Objective： To explore the anti-inflammatory effect of Anemone raddeanae alcohol extract in rats with adjuvant arthritis. Methods： Take48 SD rats were randomly divided into normal group,model group,methotrexate（ 3mg / kg） group,Anemone raddeanae alcohol extract in three different doses（ 189 mg / kg,378 mg / kg,756 mg / kg）,except for the normal group,rats in the other groups were injected Freund’s complete adjuvant into the rats’ right etapedes to establish the AA model,the rat body weight were recorded,the swelling degree of paws,arthritis index,the thymus index and spleen index were calculated,the content of IL-1,IL-6,IL-8,TNF-α and IL-10 in the rats serum were determined by ELISA method. Results： Compared with model group,the three doses of Anemone raddeanae alcohol extract can increase body weight growth trends,inhibit AA primary paws swelling of rats,reduce the secondary AA rats’ arthritis index,thymus index and spleen index,it also can cut the level of promoting inflammatory cytokines（ IL-1,IL-6,IL-8 and TNF-α） and raise the level of suppressing inflammatory cytokines（ IL-10）. Conclusion： For primary and secondary inflammation of adjuvant arthritis rats,the three doses of Anemone raddeanae alcohol extract had a significant therapeutic effect and presented a certain concentration response relationship.
