摘 要:目的:观察黄根片对四氯化碳诱导的急性肝损伤小鼠和四氯化碳复合因素诱导的肝纤维化大鼠的防治作用。方法:小鼠采用四氯化氮诱导急性肝损伤小鼠模型,灌胃给予黄根片,连续10天,观察黄根片对急性肝损伤小鼠肝脏系数,血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、抗氧化水平及肝组织病理损伤程度的影响;采用四氯化碳复合因素复制肝纤维化大鼠模型,灌胃给予黄根片,连续11周,观察黄根片对肝纤维化模型大鼠血清ALT、AST、碱性磷酸酶(ALP)、白蛋白(ALB)水平、肝脏指数、肝组织羟脯胺酸(Hyp)含量的影响,Masson染色观察肝组织纤维化程度的影响。结果:黄根片可显著降低急性肝损伤小鼠肝脏系数和ALT、AST水平,肝脏系数由模型组的(52±6)mg/g降低到黄根片36.0 g、18.0 g药材/kg剂量组的(42±6)mg/g、(45±8)mg/g,AST由模型组的(107±63)U/L降低到黄根片36.0 g、18.0 g药材/kg剂量组的(53±22)U/L、(54±31)U/L,ALT由模型组(85±41)U/L降低到黄根片36.0 g、18.0 g药材/kg剂量组的(43±16)U/L、(46±21)U/L,并可提高急性肝损伤小鼠血清、肝脏的SOD的活性,显著改善急性肝损伤小鼠肝组织病理状态。和模型组比较,灌胃给药11周,黄根片24.0g药材/kg剂量组能够显著降低大鼠肝脏指数、肝组织Hyp含量和肝纤维化百分率,分别由模型组的(8.8±1.3)g/(100g体重)、(363±96)μg/g(3.03±0.67)%下降到黄根片24.0g药材/kg剂量组的(7.1±1.4)g/(100g体重)、(244±28)μg/g肝湿重、(2.28±0.43)%肝湿重;能显著增高大鼠血清ALB水平,由模型组的(17.7±2.1)g/L肝湿重增高到黄根片24.0g原药材/kg剂量组的(19.8±1.7)g/L。结论:黄根片对四氯化碳诱导的急性肝损伤小鼠、四氯化碳复合因素所致肝纤维化大鼠均具有良好的防疗作用。Objective: To investigate the effects of huanggen tablets on acute liver injury in mice induced by carbon tetrachloride and hepatic fibrosis rats induced by carbon tetrachloride combined with high-fat and low-protein diet. Methods: After 10 days administration,acute liver injury in mice were induced by carbon tetrachloride,and the liver index,serum ALT,AST,SOD,MDA,liver tissue SOD,MDA,and the pathology change of the lever tissue were observed. Hepatic fibrosis rats were induced by carbon tetrachloride combined with high-fat and low-protein diet,after 11 weeks administration,the liver index,the degree of liver fibrosis,serum ALT,AST,ALP and ALB,Hyp in liver tissue were measured,and the severity of liver fibrosis was observed by Masson-staining. Results: Compared with model group,the liver index,huanggen tablets at 36. 0 g and 18. 0 g / kg significantly decreased the concentration of ALT and AST in serum,serum concentration of AST and ALT in model group were( 107 ± 63) U / L and( 85 ± 41) U / L,and the liver index were( 52 ± 6) mg / g. Huanggen tablets at 36. 0 g and 18. 0 g / kg,serum concentration of AST were( 53 ± 22) U / Land( 54 ± 31) U / L,serum concentration of ALT were( 43 ± 16) U / L and( 46 ± 21) U / L,the liver index were( 42 ± 6) mg/g and( 45 ± 8) mg/g,the activities of SOD were greatly increased in serum and liver homogenate( P 〈 0.05,P 〈 0. 01),and pathological results also showed that CCl4-induced liver tissue damage were improved with the treatment of huanggen tablets. Compared with model group,huanggen tablets at 24. 0 g / kg significantly decreased the liver index,the degree of liver fibrosis,and concentration of HYP in liver from( 8. 8 ± 1. 3) g /( 100g),( 363 ± 96) μg / g and( 3. 03 ± 0. 67) % of the model group to( 7. 1 ± 1. 4) g /( 100g),( 244 ± 28) ? g/g and( 2. 28 ± 0. 43) %( P 〈 0. 05,P 〈 0. 01).
