人工诱导紫穗槐多倍体条件优化研究

摘　　要：多倍体育种就是利用人工手段使作物的染色体数目加倍,用秋水仙素对植物进行多倍体诱导是人工诱导加倍的方法之一。为了探究紫穗槐多倍体诱导的最优条件,选取秋水仙素浓度、胚根长度、温度以及处理时间4个因素,用棉花蘸取秋水仙素包裹根尖诱导多倍体的发生,并对其进行形态学和细胞学的观察分析。结果表明:秋水仙素浓度0.07%,胚根长度<2 mm,温度37°C,处理时间48 h,诱导率最高。Polypoidy breeding is to double the number of chromosomes artificially.One of the methods for the polypoidy induction is to use eolchicine.Coneentration of eolchicine,length of protruded radicle,temperature and time were selected as the influencing factors to induce polypoidy of A rnorphafruticosa L.The radicle tips were covered by cotton filled with colchicine to induce polypoidy.The morphology and cytology were observerd and analysed.The orthogonal test showed the result of optimal conditions fur polypoidy induction being eolchicine concentration 0.07% ,radicle length less than 2 mm,temerature 37 ℃,and time 48 h,with which the induction rate ranked the highest.
