摘 要:目的:探究未知病因足部溃烂病人的致病因素.方法:收集10例未知病因足部溃烂病人样品,提取细菌总DNA,基于16S rDNA宏基因组学技术,测序16S rDNA基因V1~V2高变区,借助生物信息学方法分析测序数据中的菌群结构,寻找病人足部溃烂可能的致病因素.结果:10份样品共产生原始测序数据260 Mbp,优势菌门包括变形菌门、拟杆菌门、厚壁菌门、放线菌门、蓝菌门和梭杆菌门,不同样品间菌群分布有差异;同时,在属水平上统计了10份样品中均出现的细菌,其中一些与伤口的慢性感染和溃疡有关,如葡萄球菌属和链球菌属在所有样本中平均含量分别达5.37%和2.16%.结论:足部溃烂病人伤口含丰富细菌基因组,发现了伤口溃疡有关细菌线索,为揭示疾病的发生和发展提供帮助.Objective:To explore the pathogenic factors of patients with unknown etiology foot ulcers disease.Methods:10 cases foot ulcers patient samples were collected,total bacterial DNAs were extracted.Based on 16S rDNA metagenomics technology,16S rDNA gene V l~V2 highly variable region was amplified and sequenced.The flora structure of sequencing data was analyzed with bioinformatics method,looking for the suspected pathogenic factors of patients with foot ulcers.Results:Total 260 Mbp raw data in 10 samples were produced.The dominate phylum included Proteobacteria,Bacteroidetes,Firmicutes,Actinobacteria,Cyanobacteria and Fusobacteria.There were differences between samples in flora distribution.At the same time,in the genus level,bacteria appeared in ten samples were counted,some related in wounds chronic infection and ulcers,such as the average content of Staphylococcus and Streptococcus in all samples was 5.37% and 2.16% respectively.Conclusion:Bacterial genomes are rich in patients' foot ulcers wounds,bacteria related with wound ulcer were discovered,providing clues to reveal the disease occurrence and development.
