摘 要:目的研究长春市双阳区鼠类中汉坦病毒感染情况及其基因分型。方法收集2011年双阳地区捕获鼠类,取鼠肺组织,用间接免疫荧光检测,阳性标本进行汉坦病毒M片段扩增并测序,与其他毒株进行相似性比对及系统树分析。结果收集100份鼠肺,经IFA检测到1份阳性,阳性率为1%,对其进行逆转录巢式PCR扩增,结果为汉城(SEO)型HV,与代表株进行相似性比较发现,SY-4部分M基因片段序列与其他SEOV核苷酸相似性为95.4%~99.7%,氨基酸序列相似性为99%~100%。系统树显示,该序列属S3亚型。结论本次获得HV是SEO型,亚型为S3亚型。Objective To investigate the prevalence and genotypes of Hantavirus among the rodents in Shuangyang area of Changchun City. Methods The lung tissues were isolated from the rodents collected in Shuangyang District in 2011. HV antigen was detected by indirect immunofluorescence assay. Fragments of the HV M region were amplified by nested reverse transcription polymerase chain reaction( RT- PCR),sequenced and compared with other strains for similarity and phylogenetic analysis. Results A total of 100 samples were collected,of which 1( 1%)sample was positive for HV antigen. The sequencing analysis showed that the only one positive rat lung sample was SEO type of HV. The sequence alignment results of the partial M segments showed that SY- 4 held the highest similarity with other strains 95. 4%- 99. 7% nucleotide similarity and 99%- 100% amino acid similarity.Phylogenetic analysis demonstrated that this strain was grouped into the SEO subtype S3. Conclusion The study shows that the hantavirus carried by rodents in Shaungyang District Changchun City was SEOV,and the sub-genotype was SEOV- 3.
