子痫前期患者血浆miRNA表达谱的研究与分析

摘　　要：目的检测子痫前期患者和正常妊娠人群血浆中循环miRNA的表达差异,初步探讨子痫前期相关的生物学标记物。方法选取妊娠晚期的子痫前期患者8例为实验组,其中4例轻度子痫前期,4例重度子痫前期,另选正常妊娠人群8例为对照组,通过miRNA芯片技术检测实验组与对照组血浆中循环miRNA的表达情况。结果本实验共检测出15个差异miRNA,6个上调miRNA,9个下调miRNA。其中miR-584-5p在重度子痫组血浆中含量低于轻度子痫组,轻度子痫组血浆中含量低于对照组,差异有统计学意义(P<0.05)。结论循环miRNA的差异表达可能与子痫前期的发生、发展有关。miR-584-5p可能是预测子痫前期的生物学标记物。Objective To identify differentially expressed miRNAs in serum of patients with preeclampsia （PE） in comparison to unaffected controls so as to find serum biomarkers related to PE. Methods We analyzed the expression profiles of circulating miRNAs in 8 pregnant women with preeclampsia （4 cases of mild preeclampsia patient, 4 case of severe preeclampsia patients） and 8 normal controls by miRNA microarray analysis. Results In the study, 15 miRNAs were differentially expressed （6 up - regulated and 9 down - regulated） among PE cases compared with controls. Among them the levels of miRNA - 584 - 5p significantly decreased with the severity of PE （P 〈 0. 05 ）. Conclusion Those PE - related miRNAs may have a relationship with the pathogenesis and development of PE. MiRNA -584 -5p may play its potential role as a serum biomarker for forcasting PE.
