摘  要:为探讨原花青素对人胃癌SGC-7901细胞增殖及凋亡的影响及其可能的作用机制,以体外培养的SGC-7901细胞为研究对象,经一定浓度的原花青素作用后,用MTT法及流式细胞仪检测细胞增殖抑制及凋亡情况,Real-time PCR技术及免疫组化法检测Bcl-2、Bax mRNA和相关蛋白表达的含量。结果表明,不同浓度的原花青素不仅能有效抑制SGC-7901细胞增殖,还可诱导细胞凋亡,且抑制增殖及促进凋亡作用呈浓度和时间依耐性;Real-time PCR及免疫组化试验中显示,随着原花青素浓度的增加,Bcl-2mRNA及相应蛋白表达逐渐减少,Bax mRNA和相关蛋白表达逐渐增加。因此,原花青素对人胃癌SGC-7901细胞具有明显的抑制作用,其作用机制可能与上调Bcl-2蛋白和下调Bax蛋白水平有关。To investigate the anticancer effects of procyanidin on SGC-7901 cells and its possible mechanism,the SGC-7901 cell was cultured in vitro,and after a certain concentration of procyanidin treatment,the cell proliferation inhibition and apoptosis were detected by MTT assay and flow cytometry.The transcription and expression of Bcl-2,Bax were detected by immunohistochemistry and Real-time PCR.The results showed that the different concentrations of procyanidin could not only inhibit the proliferation of SGC-7901 cells,but also induce the cell apoptosis,and both of these effects were time and concentration dependent.The Real-time PCR and the immunohistochemistry test showed that the expression of Bcl-2was decreased,while the transcription and expression of Bax were increased with the increase of procyanidin concentration.The proliferation of SGC-7901 cells,as human gastric cancer cells was significantly inhibited,and the molecular mechanism may be related with up-regulation of Bax protein and down-regulation of Bcl-2protein.
