摘 要:目的:探讨脂肪源间充质干细胞(ADMSCs)静脉移植对D-半乳糖衰老模型大鼠的抗衰老效果。方法:18只SD大鼠随机平均分为3组,模型组与移植组大鼠连续8周腹腔注射D-半乳糖[100 mg/(kg·d)],对照组腹腔注射生理盐水。然后移植组尾静脉注射ADMSCs(4×106个),模型组和对照组尾静脉注射生理盐水。4周后,采用Morris水迷宫实验检测大鼠空间学习记忆能力,并测定大鼠血清中超氧化物歧化酶(SOD)活性,并用Western blot法检测大鼠肾与肺组织中衰老相关蛋白沉默信息调节因子1(Sitr1)、P16、P21及增殖细胞核抗原(PCNA)的表达。结果:与对照组相比,模型组大鼠空间学习记忆能力降低(P<0.05);与模型组相比,移植组大鼠空间学习记忆能力明显改善(P<0.05)。与对照组相比,模型组大鼠血清中SOD活性降低,肾与肺组织中Sirt1、PCNA蛋白表达水平降低(P<0.05),P21、P16蛋白表达水平升高(P<0.05);与模型组相比,移植组大鼠血清中SOD活性增加(P<0.05),肾与肺组织中Sirt1、PCNA蛋白表达水平升高(P<0.05),P21、P16蛋白表达水平降低(P<0.05)。结论:ADMSCs静脉移植治疗抗衰老作用明显,其可能通过调节衰老相关蛋白表达发挥作用。Aim: To investigate the effects of adipose-derived mesenchymal stem cells( ADMSCs) transplantation on aging in rats. Methods: A total of 18 SD rats were randomly allocated into three groups. The rats in model group and transplantation group were induced by D-galactose for 8 weeks,and control group were only injected with normal saline. And then,the transplantation group was injected with ADMSCs suspension( 4 × 10^6) by tail venous pathway,the model and control groups were injected with normal saline of the same quantity. Four weeks later,the spatial learning and memory ability of rats were detected by Morris water maze test,the serum activity of SOD was measured,and the expressions of age-related proteins in kidney and lung tissue were detected by Western blot,including silence information regulator 1( Sirt1),proliferating cell nuclear antigen( PCNA),P16 and P21. Results: Compared with control group,spatial learning and memory ability of rats in the model group reduced( P〈0. 05),while that in transplantation group was improved compared with the model group( P〈0. 05). Compared with control group,SOD activity in serum of rats in the model group significantly reduced( P〈0. 05),Sirt1 and PCNA expressions in kidney and lung tissue dramatically decreased( P〈0. 05),while P16 and P21 expressions increased( P〈0. 05). Compared with the model group,SOD activity in serum of rats in tranplantation group increased( P〈0. 05),Sirt1 and PCNA expressions increased( P〈0. 05),and P16 and P21 expressions decreased( P〈0. 05). Conclusion: ADMSCs transplantation has obviously anti-aging effect.
