摘  要:目的:通过高密度聚乙烯模具构建兔耳颗粒软骨定形移植物,并观察软骨膜对其组织学及生物力学特性的影响。方法:8只雄性新西兰大白兔,取一侧耳软骨后制备定形颗粒软骨移植物,分为保留软骨膜及去除软骨膜组,埋植于背部皮下,术后1月、2月、3月取材进行组织学及生物力学检测。结果:无论保留软骨膜与否,颗粒软骨均可长为具备一定形状和机械强度的均一整体,保留软骨膜组中可观察到Ⅱ型胶原及胶原纤维酸性蛋白(GFAP)免疫组化阳性染色的新生软骨组织,其破坏应力和弹性模量均高于去除软骨膜组(P<0.05)。结论:高密度聚乙烯模具可以很好地完成颗粒软骨定形移植物的构建,保留软骨膜的颗粒软骨具有更好的组织学活性,以及更佳的生物力学特性。Objective: To observe the effect of perichondrium on its histological and biomechanical properties by constructing diced cartilage grafts with custom shape with high-density porous polyethylene mould wrapped. Methods: Unilateral auricular cartilage were harvested from 8 rabbits and diced to lmm cube pieces, divided into 2 groups according to the preservation of perichondrium. The autologous diced cartilage grafts were implanted into the dorsal subcutaneous layer, and took out for histological and biomechanical tests at 1, 2 and 3 months respectively after the implantation. Results: The diced cartilage grafts grew into one-piece masses with certain mechanical strength in both two groups. New generative cartilage tissue with positive immunohistochemistry staining of type II collagen and glial fibrillary acidic protein (GFAP) was observed in grafts with perichondrium preserved, and their failure stress and modulus of elasticity were higher than those with perichondrium removed (P〈0.05). Conclusions: Diced cartilage graft with custom shape can be constructed with high-density porous polyethylene as mould, and the preservation of perichondrium can improve its histological and biomechanical properties.
