摘  要:目的建立HPLC-ELSD法测定胆南星中4种胆酸类成分质量分数的方法。方法色谱柱为WELCH AQ-C18柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.2%(体积分数)冰醋酸(体积比60∶40),柱温为30℃,流速为1.0 m L/min,检测器为ELSD,载气流速为2.2 m L/min,漂移管温度为90℃。结果胆酸、猪去氧胆酸、鹅去氧胆酸、去氧胆酸均在定量范围内呈良好的线性关系,平均回收率分别为97.62%、98.38%、97.32%、97.86%。结论本方法操作简单,测定结果准确,可用于胆南星中胆酸类成分的质量分数测定及质量控制。Objective To develop a HPLC-ELSD method for the determination of cholic acids from Arisaema eum Bile. Methods WELCH AQ-Cls column (4.6 mm×250 mm, 5 μm)was employed and eluted at 30 ℃ with a mobile phase consisting of aeetonitrile-0.2% glacial acetic acid ( 60 : 40), and the flow rate was 1.0 mL/min. The drift-tube temperature of ELSD was 90 ℃, and the flow rate of air was 2.2 mL/min. Results Cholie acid, hyodeoxycholic acid, ehenodeoxyeholic acid and deoxidation cholic acid had good linearity within the quantitative range, and their mean recovery ratios were 97.62%, 98.38%, 97.32% and 97.86%, respectively. Conclusion The method was simple and accurate, which could be used for the determination and quality control of cholic acid constituents from Arisaema cum Bile.
