CTGF与百草枯中毒大鼠肺纤维化相关性的实验研究

摘　　要：目的探讨结缔组织生长因子（connective tissue growth factor，CTGF）与百草枯（pa-raquat，PQ）中毒大鼠肺纤维化之间的相关性。方法将54只雄性sD大鼠随机分为对照组（NS组）、百草枯染毒组（PQ组）、法舒地尔（Fasudil）干预组（FS组）。通过给予PQ50mg／kg灌胃染毒建立百草枯中毒动物模型，Ns组给予等量生理盐水灌胃，FS组于染毒后予法舒地尔注射液30mg／（kg·d）腹腔一次注射干预，各组大鼠于第7、14、28天分批处死，采集肺组织。肺组织常规HE染色及Masson染色，显微镜下观察分析，采用Ashcroft评分法对肺组织纤维化程度进行判定，Real—timePCR检测各组肺组织CTGFmRNA的表达。结果PQ组大鼠肺组织各相应时间点Aschcoft评分、CTGFmRNA表达量均较NS组明显升高（P〈0．05），FS组大鼠肺组织各相应时间点Aschcoft评分、CTGFmRNA表达量较PQ染毒组明显降低（P〈0．05），Spearman相关分析结果显示，百草枯中毒大鼠肺组织CTGFmRNA表达与纤维化Ashcroft评分呈正相关（r=0．898，P〈0．01）。结论百草枯中毒可导致肺组织内CTGF表达明显增加，CTGF的表达量与肺纤维化程度存在显著相关性。Objective To investigate connective tissue growth factor（CTGF） in the model rats with lung fibrosis after paraquat poisoning. Methods Fifty -four male SD rats were randomly divided into three groups, control group（ NS group）, paraquat group（ PQ group）, Fasudil group（ FS group）. Rats were exposed to 50 mg/kg paraquat by oral gavage for establishing the animal model of paraquat poisoning. The rats in NS group were given equal normal saline. FS group was treated with Fasudil through intraperitoneal injection at 30 mg/（ kg ~ d）. The rats were sacrificed on 7,14,28 days after poinsoning. The lung tissue was harvested to HE staining and Masson staining. Lung fibrosis was assessed by Ashcroft score. Real - time PCR was used to test the content of CTGF mRNA in the lung tissues of the rats of three groups. Results The Ashcroft score of lung tissue and CTGF mRNA in lung of PQ group were higher than those in NS group（P 〈 0.05 ）. While the Ashcroft score of lung tissue and CTGF mRNA in lung of PQ group were lower than those in PQ group （ P 〈 0. 05 ）. The CTGF mRNA was associated with increased Ashcroft scores （ Spearman r = 0. 089, P 〈 0.01 ）. Conclusion CTGF expression is increased significantly in the lung tissues after paraquat poisoning. The expression of CTGF is related with the degree of lung fibrosis.
